摘要 目的分析黑龙江产地甜瓜籽中脂肪酸的组成,建立同时测定甜瓜籽中棕榈酸、硬脂酸、油酸和亚油酸4种脂肪酸成分的方法。方法采用GC-MS法确证甜瓜籽中脂肪酸的组成,GC法测定甲酯化后甜瓜籽油中棕榈酸、硬脂酸、油酸和亚油酸的含量。结果甜瓜籽中含7种脂肪酸,棕榈酸、硬脂酸、油酸和亚油酸的线性分别为83.2~665.6、42.8~342.4、48.4~387.2、262.0~2 096.0 mg·L-1,相关系数分别为0.999 9、0.999 3、0.999 1、0.999 0,平均回收率分别为(99.3±1.18)%、(100.8±1.47)%、(101.4±1.47)%、(101.3±1.26)%,RSD分别为1.19%、1.46%、1.45%、1.24%(n=9)。结论鉴定了甜瓜籽中7种脂肪酸组成,首次建立了甜瓜籽中4种主要脂肪酸的含量测定方法,可为甜瓜籽的质量控制提供依据。 Objective To analyze the composition of fatty acids in muskmelon seed and to develop a method for the determination of hexadecanoic acid,stearic acid,oleic acid and linoleic acid in muskmelon seed.Methods After methyl esterification,the composition of fatty acids were identified by GC-MS,and the contents of 4 kinds of main fatty acids were determined by GC. Results 7 kinds of fatty acids in muskmelon seed were confirmed. The calibration curves were linear within the range of 83. 2- 665. 6,42. 8- 342. 4,48. 4-387. 2 mg·L- 1and 262. 0- 2 096. 0 mg·L- 1and the correlation coefficients were 0. 999 9,0. 999 3,0. 9991 and 0. 999 0 for palmic acid,octadecanoic acid,loeic acid and linoleic acid respectively. The average recoveries( n = 9) were( 99. 3 ± 1. 18) %,( 100. 8 ± 1. 47) %,( 101. 4 ± 1. 47) % and( 101. 3 ± 1. 26) % for hexadecanoic acid,stearic acid,oleic acid and linoleic acid respectively. Conclusions Seven kinds of fatty acids in muskmelon seed are identified and a method for the determination of four kinds of fatty acids simultaneously is developed. The method has good sensitivity,accuracy and repeatability and can be used for the quality control of the muskmelon seed.
