青蒿酸对人肝癌细胞SMMC-7721细胞增殖抑制的体外研究 被引量:2

Inhibitory Effects of Artemisinic Acid on Proliferation of Human Liver Cancer SMMC-7721 Cells in Vitro

摘要 目的探讨青蒿酸体外作用对人肝癌细胞SMMC-7721细胞增殖的影响及其机制。方法采用MTT法检测青蒿酸对SMMC-7721细胞增殖的影响,Hoechst-33258、Annexin V-FITC/PI检测细胞凋亡情况。结果青蒿酸作用SMMC-7721细胞24,48,72 h,能显著抑制细胞增殖,并呈时间、剂量依赖性。青蒿酸作用48 h后,SMMC-7721呈明显的凋亡形态,高浓度青蒿酸作用48 h后,呈典型的细胞凋亡特征,凋亡率达67.54%。结论青蒿酸能抑制SMMC-7721细胞增殖,这可能和诱导细胞凋亡有关。 Objective To investigate the inhibition of artemisinie acid on the proliferation of human liver SMMC-7721 cells and to explore its mechanism in vitro. Methods MTF assay was used to observe the effects of artemisinic acid on the proliferation of SMMC-7721 cells. Apoptosis of the cells was analyzed after staining with Hoechest-33258 and Annexin V-FITC/PI. Results The results of MTT assay showed that artemisinie acid could significantly inhibit the proliferation of SMMC-7721 cells after 24, 48 and 72 hours, and the effects were in time- and concentration- dependent manner. Hoechest-33258 staining results presented obvious apoptotic morphology in SMMC-7721 cells, and the results of Annexin V-FITC/PI double-staining indicated that the apoptotic rate of 5MMC-7721 cells arrived to 67.54% after treated with high-concentration artemisinic acid. Conclusion Artemisinic acid can inhibit the proliferation of SMMC-7721 cells, which may be related to the induction of apoptosis of tumor cells.
