摘要
目的:研究盐霉素对头颈部鳞状细胞癌(HNSCC)Fadu细胞增殖和侵袭的影响,初步探讨盐霉素在HNSCC发生发展中的作用。方法:不同浓度盐霉素(2、4、8、16μMmol/L)作用于Fadu细胞,分别处理12、24、36、48 h,用MTT法检测盐霉素对Fadu细胞增殖的影响,Transwell小室法测定盐霉素对Fadu细胞迁移和侵袭能力的影响;Western blot法检测盐霉素对Fadu细胞中基质金属蛋白酶-7(MMP-7)、基质金属蛋白酶-9(MMP-9)和基质金属蛋白酶-13(MMP-13)蛋白表达的影响。结果:盐霉素呈时间-剂量依赖性抑制Fadu细胞的增殖(P<0.05),使Fadu细胞的迁移和侵袭能力明显下降。盐霉素还使MMP-7和MMP-9蛋白的表达水平下调(P<0.05)。结论:盐霉素可以降低Fadu细胞的增殖能力,抑制该肿瘤细胞的迁移和侵袭能力,迁移和侵袭能力的改变可能与下调MMP-7和MMP-9的蛋白表达有关。
Objective:To investigate the effects of salinomycin on the biological behaviors of squamous cell carcinoma of the Hypopharyngeal Fadu cells.Method:The Fadu cells were treated with different concentrations of salinomycin(2,4,8and 16 μMmol/L) for 12,24,36 and 48 h.The effect of salinomycin on proliferation of Fadu cells was measured by using MTT assay.Migration and invasion of Fadu cells were determined by Transwell assay.The protein expressions of MMP-7,MMP-9 and MMP-13 were evaluated by Western blot.Result:The proliferation of Fadu cells were significantly reduced by salinomycin treatment in a time-and dose-dependent manner(P〈0.05).Compared with that in the control group,the ability of migratory and invasive cells was remarkably decreased(P〈0.05).In addition,down-regulation of MMP-7 and MMP-9were observed.Conclusion:These results demonstrated that salinnomycin can effectively inhibit proliferation,migration and invasion of Fadu cells in vitro,probably via the down-regulating the protein expressions of MMP7 and MMP-9.
