摘要
转基因鸡作为重要的经济动物和理想的生物反应器,具有广阔的应用前景。鸡基因组测序的完成加快了转基因鸡的研究和发展。病毒载体法是转基因鸡制备的主导方法之一,具有效率高和遗传稳定性良好的优势。本文对逆转录病毒载体特征、复制缺陷型病毒载体和复制整合双缺陷型病毒载体的优缺点及其介导的转基因鸡研究进行综述,展望人工核酸酶技术结合复制整合双缺陷型逆转录病毒载体在转基因鸡研究领域的应用前景。
Transgenic chicken is one of the most important economic animals and ideal bioreactors. The accomplishment of chicken genome sequencing promotes the development of transgenic chicken research. Taking the advantage of high efficiency and genetic stability,viral vector method is one of the primary strategies to generate transgenic chicken. Hereby,characteristics of retrovirus,replication defective retroviral vector and replication-integration double defective retroviral vector,and their application in transgenic chicken studies were reviewed. In addition,combining artificial nucleases with replication-integration double defective retroviral vector could be applied in the era of transgenic chicken in near future,which could provide useful information to other researchers.
