摘要
目的 :探讨成纤维细胞 (FB)与细胞间粘附分子_1(ICAM_1)在口腔粘膜下纤维化 (OSF)发生中的可能作用。方法 :分别从正常 (NM)及OSF患者的口腔粘膜中培养出FB ,向培养基中加入不同浓度的槟榔提取物 (ANE)培养 4 8h后 ,用细胞酶联免疫方法检测FB表达的ICAM_1水平。结果 :表示ICAM_1水平高低的OD值在OSF_FB为 0 386±0 0 99,高于NM_FB的OD值 0 32 4± 0 0 30 (P <0 0 5 ) ;ANE在 5 0~ 15 0 μg/ml的范围内以浓度—效应依赖关系刺激FB产生ICAM_1。结论 :ANE能上调FB表达ICAM_1的水平 ,提示ICAM_1及其介导的细胞与细胞或细胞与基质间的相互作用在OSF的发生、发展中可能起重要作用。
Objective:The study aimed at investigating the effects of areca nut extracts on the levels of ICAM_1 expressed by the oral fibroblasts.Methods: The fibroblasts were obtained from normal buccal mucosa and buccal mucosal with submucous fibrosis and cultured in vitro. The levels of ICAM_1 expressed by fibroblasts incubated with/without ANE in the presence of 10% fetal calf serum in the mixed gas of 5% CO\-2 and air at 37℃ for 48 h, were monitored using cell_based ELSA.Results: The fibroblasts obtained from mucosal with oral submucous fibrosis expressed ICAM_1 at a high level. ANE increased the expression levels of ICAM_1 in both fibroblasts obtained from normal and abnormal mucosa.Conclusion: The high expression levels of ICAM_1 in the fibroblasts of oral submucous fibrosis and ANE stimulated the expression of ICAM_1, which may be one of the important mechanisms why lymphocytes become localized and retained at the side of connective tissue during inflammation and lead to activation of fibroblasts.\;
