摘要
目的观察姜黄素(Cur)对大鼠体外循环(CPB)所致急性肺损伤(ALI)的影响。方法实验用SD大鼠50只,按照随机数字表法分为5组(n=10):假手术组(Sham组)、CPB组、CPB+Cur剂量分别为100、150、200mg/kg组(Cur.100组、Cur.150组和Cur-200组)。CPB结束后留取左肺,测定肺湿/干重比(W/D)和总肺水含量(TLW),光镜下测定肺泡损伤数定量评价指标(IQA),电镜下观察肺组织超微结构改变,反转录.聚合酶链反应(RT.PCR)和Westernblot分别检测天冬氨酸特异性半胱氨酸蛋白酶(Caspase).12mRNA及蛋白表达水平,原位末端细胞凋亡检测法(TUNEL法)检测肺组织细胞凋亡指数(AI)。结果与Sham组比较,CPB组Caspase-12mRNA水平(0.96±0.18)及蛋白表达水平(2.79±0.74)均升高(P〈O.05),W/D(4.99±0.72)、TLW(3.99±0.72)、IQA(40.22±5.39)和AI(35.19±5.94)均增加(P〈0.05),肺组织超微结构发生明显损伤;与CPB组比较,Cur-100组、Cur-150组和Cur-200组Caspase-12mRNA水平(0.87±0.18、0.69±0.13、0.56±0.12)及蛋白表达水平(2.88±0.79、1.96±0.58、1.34±0.49)均降低(P〈0.05),W/D(4.13±0.89、3.43±0.97、2.65±0.85)、TLW(3.13±0.89、2.43±0.97、1.65±0.85)、[QA(31.47±3.48、20.52±2.69、14.99±3.56)和AI(31.49±2.91、24.78±3.41、14.47±2.69)均下降(P〈0.05),肺组织超微结构损伤均有不同程度的减轻,以Cur-200组上述指标改善最为显著。结论200mgZkg的Cur对CPB引起的大鼠肺损伤具有较好的保护作用,其机制可能与其抑制Caspase-12表达,减少细胞凋亡有关。
Objective To investigate the effect of curcumin (Cur) on acute lung injury (ALI) in- duced by cardiopulmonary bypass (CPB) in rats. Methods Fifty Sprague -Dawley (SD) rats were ran- domly allocated into five groups (n = 10) : Sham operation group (Sham group), CPB group, CPB + Cur pre -treated with respectively 100, 150 and 200 mg/kg groups (Cur - 100 group, Cur - 150 group and Cur-200 group). Rats were euthanized after CPB, and left lung tissue was excised. Wet lung weight to dry lung weight (W/D) and total lung water content (TLW) were tested. Index of quantitative evaluation for alveolar damage (IQA) was tested by light microscope. Pathological changes of ultrastructure of lung lissue were observed by transmission electron microscope. The expression levels of cycteinyl aspirate - spe- cific protease (Caspase) - 12 mRNA and protein were detected respectively by reverse transcription - poly- merase chain reaction ( RT - PCR) and Western blotting. Pneumocyte apoptosis index (AI) was deter- mined by terminal dexynucleotidyl transferase (TdT) -mediated dUTP nick end labeling (TUNEL) meth- od. Results Compared to Sham group, the expression levels of Caspase - 12 mRNA (0. 96 ± 0. 18 ) and protein (2. 79 ± 0. 74 ) were all significantly increased ( P 〈 0. 05 ) in CPB group, and W/D ( 4. 99 ± 0. 72 ), TLW ( 3.99 ±0. 72), IQA ( 40. 22 ±5.39 ), AI ( 35.19 ±5.94) were all notably higher ( all P 〈 0. 05 ) ; Uhrastructural injury in lung tissue was notably observed in CPB group. Compared to CPB group, the expression levels of Caspase - 12 mRNA (0. 87 ± 0. 18, 0. 69 ±0. 13, 0. 56 ±0. 12) and protein ( 2. 88 ±0. 79, 1.96±0. 58, 1.34 ±0. 49)were decreasingly lower (P 〈0. 05) in Cur - 100 group, Cur - 150 group and Cur -200 group, W/D (4. 13 ±0. 89, 3.43 ±0. 97, 2. 65 ±0. 85), TLW (3. 13 ±0. 89, 2. 43 ±0. 97, 1.65 ±0.85), IQA (31.47±3.48, 20.52 ±2.69, 14.99 ±3.56), AI (31.49 ±2.91, 24.78 ±3.41, 14. 47 ± 2. 69 ) were also decreased ( all P 〈 0. 05 ) ; Ultrastruetural injury in lung tissue were gradually al-leviated in Cur - 100 group, Cur - 150 group and Cur - 200 group. Conclusion Cur at a dose of 200 mg/kg has protective effects on lung against injury induced by CPB in rats, which may be related to in- hibition of pneumocyte apoptosis mediated by Caspase - 12.
