咳喘宁含药血清对呼吸道合胞病毒诱发大鼠哮喘模型CD_4^+T、CD_8^+T细胞内PI3K、STAT6的干预研究 被引量:9

摘要 目的:探讨咳喘宁含药血清对呼吸道合胞病毒诱发大鼠哮喘模型CD4+T、CD8+T细胞内PI3K、STAT6的干预作用。方法:制作呼吸道合胞病毒诱发大鼠哮喘模型,提取并培养CD4+T、CD8+T细胞,采用RT-PCR和WesternBlot方法观察咳喘宁含药血清对呼吸道合胞病毒诱发大鼠哮喘模型CD4+T、CD8+T细胞内PI3K、STAT6的干预作用。结果:咳喘宁组PI3K m RNA、STAT6 m RNA表达量均较模型组和阳性药组降低,差异有统计学意义(P<0.05)。咳喘宁组PI3K蛋白、STAT6蛋白的相对表达量均较模型组和阳性药组降低,差异有统计学意义(P<0.05)。结论:咳喘宁含药血清对呼吸道合胞病毒诱发哮喘模型大鼠CD4+T、CD8+T细胞内PI3K、STAT6及其m RNA过高的表达产生明显抑制作用,从而纠正Th2细胞的过度增殖分化,影响Th2免疫应答优势,可能为其防治哮喘的作用机制之一。 Objective: To investigate the intervention effect of serum containing methoxyphenamini hydrochloridum on expression of PI3K and STAT6 in CD:T and CD+T cells of asthma rat model induced by respiratory syncytial virus. Methods: Asthma rat model was established by using respiratory syncytiai virus. The intervention effect of serum containing methoxyphenamini hydrochloridum on expression of PI3K and STAT6 in CD:T and CD+T cells of asthma rat model induced by respiratory syncytial virus were observed by using RT-PCR and Western-blotting methods. Results: The mRNA expression of PI3K and STAT6 of rats in methoxypbenamini hydrochloridum group was decreased significantly compared with rats in model group and control group, and the difference among these groups has statistical significance (P〈0.05). The protein expression of PI3K and STAT6 of rats in methoxyphenamini hydrochloridum group was decreased significantly compared with rats in model group and positive control group, and the difference among these groups has statistical significance (P〈0.05). Conclusion: Serum containing methoxyphenamini hydrochloridum could inhibit the protein and rnRNA expression of PI3K and STAT6 in CD4+T and CD8+T cells of asthma rat model induced by respiratory syncytial virus obviously, and influence the immune response of Th2 cells by correcting the excessive proliferation and differentiation of Th2 ceils, which might be one of the mechanisms of preventing and treating asthma with methoxyphenamini hydrochloridum.
