摘要
目的:观察依据六经辨证之三阴病,运用扶阳护阴、运转枢机法对糖调节受损(Impaired glucose regulation,IGR)大鼠血清氧化应激状态的影响。方法:采用高热量饮食加小剂量链脲佐菌素(Streptozotocin,STZ)注射法制作IGR大鼠模型。8只正常大鼠作为正常组,造模成功的24只IGR大鼠随机分为模型组、中药组、二甲双胍组,每组8只大鼠。观察各组大鼠4周后血清氧化应激状态与抗氧化酶水平,包括血清超氧化物歧化酶(Superoxide dismutase,SOD)、丙二醛(Malondialdehyde,MDA)、过氧化氢酶(Catalase,CAT)的检测及血清总抗氧化能力(Total antioxidant capacity,T-AOC)、谷胱甘肽过氧物酶(Glutathione peroxidase,GSH-PX)、维生素C(Vitamin C,VC)、一氧化氮(Nitric oxide,NO)、一氧化氮合酶(Nitric oxide synthase,NOS)的活力或含量等指标的测定。结果:中药组T-AOC含量升高,MDA含量降低(P<0.05),与二甲双胍组无显著性差异(P>0.05)。中药组SOD、CAT、GSH-PX活力、VC水平升高,NOS降低(P<0.05),二甲双胍组与模型组对比无显著性差异(P>0.05)。中药组NO降低(P<0.05),与二甲双胍组有显著性差异(P<0.05)。结论:从六经辨证之三阴病入手,扶阳护阴、运转枢机法中药复方可明显减轻脂质过氧化,升高抗氧化酶含量与活性,疗效优于二甲双胍。
Objective: To observe the effect of strengthening Yang, protecting Yin and circumrotating Shuji (SPC) method on oxidative stress in IGR rats. Methods: The IGR rats were made by high fat feeding and injecting small dose STZ. Eight normal SD rats were used as control group. Twenty four IGR rats were randomly divided into model group, TCM group and metformin group, 8 rats for each group. The course of treatment lasted 4 weeks. The effects on the index of oxidative stress were observed, for example SOD, MDA, CAT, GSH-Px and so on. Results: In TCM group, the level of T-AOC was increased, while MDA was decreased (P〈0.05), compared with metformin group, there were no difference (P〉0.05). The activity of SOD, CAT, GSH-PX, VC were increased and NOS was decreased in TCM group (P〈0.05), while there were no effects on those in metformin group. The level of NO in TCM group decreased (P〈0.05) and the effect was significantly better than metformin group (P〈0.05). Conclusions: Strengthening Yang, protecting Yin and circumrotating Shuji method can improve oxidative stress availably, it can reduce over-oxidation of lipid, elevating the content and activeness of anti-oxidase, the curative effect suroasses metformin.
