摘要
目的研究水蒸汽蒸馏法提取枫香脂挥发油对大鼠离体胸主动脉的舒张作用,并探讨其可能机制。方法枫香脂挥发油成分分析在气相-质谱上进行,分离SD大鼠胸主动脉进行离体血管环实验,观察枫香脂挥发油对苯肾上腺素预收缩引起的舒缩功能的反应,并结合一氧化氮合酶抑制剂左旋硝基精氨酸甲酯和环氧合酶抑制剂吲哚美辛,观测枫香脂挥发油对血管环的作用。结果气相-质谱结果显示主要成分是三甲基肼、3-蒈烯、(1R)-2,6,6-三甲基二环[3.1.1]庚-2-烯、2-亚甲基-4,8,8-三甲基-4-乙烯基-二环[5.2.0]壬烷;累计体积分数的枫香脂挥发油对苯肾上腺素预收缩的血管环在内皮完整最大舒张率可达32.67%;结合左旋硝基精氨酸甲酯和吲哚美辛孵育0.5 h后,均显示有明显抑制血管舒张的作用。结论枫香脂挥发油对大鼠离体血管确实有舒张血管的作用,其机制可能跟一氧化氮合酶和环氧合酶的合成有关。
AIM To investigate the vasodilatory effect of Liquidambaris Resina steam-volatile oil on the isolated thoracica aorta and explore its possible mechanism. METHODS The analysis of Liquidambaris Resina volatile oil was performed on gas chromatography-mass spectrometry. Isolated SD rat aortic vascular was used in vitro to the vascular ring experiment,observing the pre-contraction effect caused by phenylephrine,as well as by nitric oxide synthase inhibitor L-NAME and cyclooxygenase inhibitor indomethacin on vascular ring. RESULTS GC-MS data showed that the main constituents of volatile oil were trimethyl-hydrazine,3-carene,( 1R)-2,6,6-trimethylbicyclo [3. 1. 1] hept-2-ene,2-methylene-4,8,8-trimethyl-4-vinyl-bicyclo-[5. 2. 0] nonane. Volatile oil had a significant pre-contracted effect caused by PE,and the value of maximum relaxation was 32. 67%. After L-NAME and indomethacin incubating with vascular ring for 0. 5 h,they could markedly inhibit vasodilation. CONCLUSION Liquidambaris Resina volatile oil has significant vasodilation for SD rats,and is mainly related to the activation of nitric oxide synthase and cyclooxygenase pathway.
