摘要
目的观察磷酸川芎嗪(LP)贴剂对大鼠急性心肌缺血的保护作用及凝血时间的影响。方法采用舌下静脉注射垂体后叶素(Pit)致大鼠急性心肌缺血模型,将大鼠随机分为模型组、赋形剂组、阳性组及LP贴剂高、中、低(4、2、1 cm2)剂量组等6组。观察各组大鼠注射Pit后不同时间点的Ⅱ导联心电图S-T段及T波的变化情况。采用毛细管法于眼眶取血,测定大鼠的凝血时间。观察LP贴剂3个剂量组与赋形剂组对大鼠凝血时间的影响。结果 LP贴剂高、中、低剂量组均能明显抑制注射Pit后15 s^5 min内大鼠心电图S-T段与T波的变化,均能明显延长大鼠的凝血时间。结论 LP贴剂对Pit致大鼠急性心肌缺血有明显的保护作用,具有抗凝血即抑制血小板聚集的作用。
OBJECTIVE To investigate the protective effect of Ligustrazine phosphate(LP) patches on the myocardial ischemia rats and the effect on blood clotting time of rats. METHODS The animal model of myocardial ischemia induced by injecting Pituitrin (Pit) through sublingual vein was built to evaluate the efficacy of LP patches against myocardial ischemia in rats. The rats were divided into 6 groups randomly involving model control group, excipient group, positive control group, patches of low - dose group, patches of medium - dose group and patches of high - dose group ( 1,2,4 cm2 ). The migration of S - T period and abnormality of T wave at the different time points of standard Ⅱ lead electrocardiogram (ECG) in rats of myocardial ischemia were observed. The blood clotting time was surveyed by the method of capillary. The blood clotting time in rats of the three doses of LP groups and the excipient control group was evaluated. RESULTS The displacements of S - T period and T wave in 15 s - 5 min of ECG could be improved obviously after 3 doses of LP patches were administrated on acute myocardial ischemia the rats caused by Pit. The blood clotting time could be extended after 3 doses of LP patches were administrated on rats. CONCLUSION LP patches can protect the myocardial ischemia rats obvious- ly. LP patches have the anticoagulation activity.
