摘要
目的探讨趋化因子CCL27、CCL28及其受体CCR10在小鼠癫痫急性期外周血中的表达变化。方法取正常及癫痫小鼠急性期不同时间点(10min、30min、1h、2h)外周血利用实时荧光定量PCR(Real-time PCR)法检测毛果云香碱致癫痫发作小鼠急性期外周血中CCL27、CCL28mRNA水平;同期取正常及癫痫急性期不同时间点(10min、30min、1h、2h)肝素抗凝外周血,提取淋巴细胞层后利用FACSsort型流式细胞仪检测外周血淋巴细胞CCR10蛋白水平的表达变化。结果癫痫小鼠发作急性期外周血中CCL27、CCL28mRNA水平、淋巴细胞中CCR10蛋白表达水平均在癫痫发作2h高于对照组(P<0.05)。结论癫痫发作早期已出现外周血免疫功能紊乱,该病理变化可能与癫痫发病过程中的炎性反应和神经细胞的凋亡相关。
Objective To investigate the expression of chemokine CCL27,CCL28 and their receptor CCR10 in mouse periph- eral blood in the acute phase of epilepsy. Methods The peripheral blood of acute epileptic mice at different time points(10 rain,30 min, 1 h,2 h) was collected,real-time PCR was used to detect the mRNA expression level of CCL27 and CCL28. The heparin anti- coagulation peripheral blood at the same time points(10 min, 30 min, 1 h, 2 h) of normal and acute phase of epileptic mice were col- lected and flow cytometry was used to investigate the expression of CCR10 in peripheral blood lymphocyte. Results The mRNA expression level of CCL27, CCL28 in peripheral blood and the expression of CCR10 in lymphocytes were found significantly in- creased at 2 h in epileptic mice than those of normal(P〈0.01). Conclmion The immune function disorder oecured in peripheral blood in early epilepsic pathological process and might be associated with the subsequent inflammatory reaction and neuron apoptosis.
