摘要
建立了一种测定菊花中7种单体酚含量的超高效液相色谱法。采用ACQUITY UPLC HSS T3色谱柱,以甲醇/乙酸铵-乙酸缓冲溶液为流动相,梯度洗脱,变波长扫描,各化合物线性较好(R2〉0.993),在不同的添加水平都得到了较满意的回收率(于87.0%~109.0%之间)和较好的方法重复性(RSD≤6.25%)。测定了三种名菊和新疆雪菊中7种单体酚的含量,结果表明,四种菊花中含有的单体酚种类不同,含量也有差异,其中儿茶素和槲皮素的含量都很丰富。
A UPLC method was developed for determining the contents of 7 mono-phenols in chrymnflaemum. The chromatographic separation was performed on a ACQUITY UPLC HSS T3 column at 30 ℃ with methanol/ammonium acetate-acetic acid mixture as a mobile phase at a flow rate of 0.2 mL/min. The detection wavelength was variable with the change of compounds. The calibration curves of 7 mono-phenols were linear with correlation coefficients more than 0.993, and recovery (87.0%--109.0%) and accuracy (RSD ≤ 6.25%) of every compound was satisfactory. The content of 7 mono-phenols was analyzed in Xinjiang Coreopsis tinctoria and other three chrysanthemum. The results showed that different mono-phenol and unequal amount were found, and rich catechin and quercetin were detected in four chrysanthemum.
