摘要
目的探讨肿瘤低表达长链非编码RNA(lncRNA-LET)基因对食管鳞癌Eca109与TE-1细胞的增殖、凋亡、侵袭和迁移能力的影响。方法 RT-PCR检测36例食管鳞癌患者癌组织及癌旁组织中lncRNA-LET的表达水平。采用慢病毒转染Eca109与TE-1细胞过表达lncRNA-LET。比较转染后(Eca109LET组和TE-1LET组)细胞与正常培养(Eca109组和TE-1组)细胞的增殖、凋亡、侵袭和迁移能力的变化。结果 LncRNA-LET在食管鳞癌组织中的表达低于癌旁组织(P<0.05)。Eca109LET组和TE-1LET组细胞每个视野的穿膜数量、细胞的迁移距离、细胞存活率分别低于Eca109组和TE-1组(P<0.05),而发生凋亡的细胞比例分别高于Eca109组和TE-1组(P<0.05)。结论 LncRNA-LET基因在食管鳞癌组织中低表达;其过表达能够抑制食管鳞癌Eca109和TE-1细胞的增殖、侵袭和迁移能力,并能促进其凋亡。
Objective To investigate the effects of lncRNA-low expression in tumor(lncRNA- LET) gene transfection on the proliferation, apoptosis, invasion and migration of esophageal squamous carcinoma(ESCC) Ecal09 and TE-1 cell lines. Methods The expression of lncRNA-LET mRNA in primary and matched tumor-adjacent normal tissues of 36 patients was determined by RT-PCR. Lentivirus infection technique was employed to up-regulate the expression of lncRNA-LET in Ecal09 and TE-1 cell lines. The cell proliferation, apoptosis, invasion and migration were measured and compared between transfected cells(group Ecal09 LET and group TE-1 LET) and cultured normal cells(group Ecal09 and group TE-1). Results The expression of lncRNA-LET was lower in primary ESCC tissues than that in tumor-adjacent normal tissues(P〈0. 05). The number of wear membrane cells, cell migration distance and cell survival rate were all lower, and apoptosis cell ratio was higher, in group Ecal09 LET and group TE-1 LET than those in group Ecal09 and group TE-1, respectively (P〈0. 05). Conclusion LncRNA-LET gene expression in primary ESCC tissues is lower, and over- expression of lncRNA-LET can inhibit the proliferation, invasion and migration and promote the apoptosis of the ESCC Eca109 and TE-1 cells.
