摘要
目的探讨17B-雌二醇在晶状体氧化损伤中的保护作用。方法 50只雌性大鼠随机分为5组:正常对照组、模型组、卵巢切除组、卵巢切除+保护组、假手术组,每组10只。除正常对照组外,其他4组制作眼前段紫外线损伤模型。卵巢切除组给予苯甲醇,花生油;卵巢切除+保护组切除卵巢4周后给予17p-雌二醇,0.5蚓d,连续3 d;假手术组开腹找到双侧卵巢后即关腹。取大鼠晶状体囊膜检测过氧化产物丙二醛(MDA)含量和超氧化物歧化酶(sOD)活性,流式细胞仪检测晶状体上皮细胞凋亡率,在电镜下观察凋亡细胞超微机构。结果模型组、卵巢切除+保护组、假手术组:MDA含量和晶状体上皮细胞凋亡率较卵巢切除组低(P<0.05),而3组之间MDA含量和晶状体上皮细胞凋亡率差异无统计学意义。正常对照组MDA含量及晶状体上皮细胞凋亡率较其他4组均低(P<0.05)。正常对照组SOD活性高于其他4组,模型组、卵巢切除+保护组、假手术组SOD活性较卵巢切除组高(P<0.05),而3组之间差异无统计学意义。透射电镜下凋亡细胞呈现体积缩小,染色质边集、浓缩的典型改变。结论紫外线诱导晶状体蛋白发生氧化损伤,使晶状体上皮细胞发生凋亡,从而导致白内障形成。17D-雌二醇有抗氧化、防止白内障形成的保护作用。
Objective To investigate the protective effects of 17β-estradiol on oxidative damage in the lens. Methods Fifty femal Wistar rats were randomly divided into five groups: normal group, model group, ovariectomy group, ovariectomy with estrodiol replacement therapy group, sham group. Damage model of anterior segment of eyes by ultraviolet (UV) was induced in each group except normal group. In ovariectomy with estrodiol replacement therapy group, the rats were given injections of 0.5 μg 17β-estradiol (dissolved in benzyl alcohol (1:3, vol/vol) daily for three days four weeks after ovariectomy, while ovariectomy group received vehicle alone. The bilateral ovary was found and the abdomen wall was closed in sham group. Contents of malo- ndialdehyde and activities of superoxide dismutase of tissues surrounding ocular of rats lenses were measured and the apoptosis rate of lens epithelial cells of previous lens capsule was also detected by flowcytometry. Moreover, the super microstructures of apoptotic cells were observed with TEM. Results The malonaldehyde (MDA) level and lens epithelial cells apoptosis rate in ovariectomy group were significantly higher compared with model group, ovariectomy with estrodiol replacement and sham group (P 〈 0.05), no differences were found among model group, ovariectomy with estrodiol replacement and sham groups. The level of MDA and the apoptosis rate of lens epithelial cells in normal group were lower than the other groups (P 〈 0.05). The levels of superoxide dismutase in normal group were higher than other groups (P 〈 0.05), that in ovariectomy group were lower than that in model, ovariectomy with estrodiol replacement therapy and sham operated groups other groups (P 〈 0.05), among which no significant difference was found. In addition, typical characteristic changes of cell apoptosis, the chromatin edging and condensing were observed by TEM. Conclusion UV could induced oxidative damage in lens protein and apoptosis of epithelial cells, resulting in the formation of cataracts. Estrogen has anti-oxidant protection and may appears a good alternative clinical option for cataract prevention and treatment.
