六味地黄丸降低手机频率电磁辐射对大鼠睾丸组织的损伤 被引量:16

Protective effect of Liuweidihuang Pills against cellphone electromagnetic radiation-induced histomorphological abnormality,oxidative injury,and cell apoptosis in rat testes

摘要 目的:观察六味地黄丸对手机频率电磁辐射雄性大鼠睾丸组织形态学、氧化损伤和细胞凋亡的影响。方法:30只雄性SD大鼠随机分为正常组、辐射组、六味地黄丸组,每组各10只。正常组不辐射,辐射组和六味地黄丸组大鼠每天暴露于900 MHz手机频率辐射4 h共18 d;六味地黄丸组大鼠每日灌胃六味地黄丸悬液(1 ml/100 g体重),其他组大鼠每天灌胃等量纯净水。实验结束后腹腔麻醉取材处死大鼠,分别取睾丸组织固定和冷冻,常规HE染色观察组织形态学变化,比色法检测睾丸组织丙二醛(MDA)、谷胱甘肽(GSH)水平,免疫组化法观察睾丸细胞bax,bcl-2蛋白表达的平均光度值。结果:与正常组相比,辐射组大鼠睾丸生精小管细胞结构松散,精母细胞层数减少,内有空洞出现;睾丸组织MDA水平显著升高[(0.36±0.07)μmol/g prot vs(0.25±0.06)μmol/g prot,P<0.05],GSH水平显著降低[(42.10±5.08)mg/g prot vs(52.29±4.16)mg/g prot,P<0.05];辐射组大鼠睾丸生精细胞bax蛋白表达显著增强(0.91±0.10 vs 0.21±0.01,P<0.01),bcl-2蛋白表达显著减弱(0.18±0.07 vs 0.70±0.94,P<0.01)。六味地黄丸组大鼠睾丸组织形态学接近正常,与辐射组比较,睾丸组织MDA含量显著降低[(0.30±0.05)μmol/g prot,P<0.05],GSH含量无明显变化[(45.34±5.21)mg/g prot,P>0.05],睾丸生精细胞bax蛋白表达(0.41±0.05)明显减弱(P<0.01),bcl-2蛋白表达(0.27±0.08)明显增强(P<0.01)。结论:六味地黄丸可改善手机频率电磁辐射造成的大鼠睾丸组织结构损伤,降低睾丸组织氧化损伤,减少睾丸细胞凋亡。 Objective: To observe the effect of Liuweidihuang Pills in relieving cellphone electromagnetic radiation-induced histomorphological abnormality, oxidative injury, and cell apoptosis in the rat testis. Methods: Thirty adult male SD rats were equal- ly randomized into a normal, a radiated, and a Liuweidihuang group, the animals in the latter two groups exposed to electromagnetic radiation of 900 MHz cellphone frequency 4 hours a day for 18 days. Meanwhile, the rats in the Liuweidihuang group were treated with the suspension of Liuweidihuang Pills at 1 ml/100 g body weight and the other rats intragastrically with the equal volume of purified wa- ter. Then all the rats were killed for observation of testicular histomorphology by routine HE staining, measurement of testicular malon- dialdehyde (MDA) and glutathione (GSH) levels by colorimetry, and determination of the expressions of bax and bcl-2 proteins in the testis tissue by immunohistochemistry. Results: Compared with the normal controls, the radiated rats showed obviously loose struc- ture, reduced layers of spermatocytes, and cavitation in the seminiferous tubules. Significant increases were observed in the MDA level ( P 〈 0.01 ) and bax expression ( P 〈 0.01 ) but decreases in the GSH level ( P 〈 0.01 ) and bd-2 expression ( P 〈 0.01 ) in the testis tissue of the radiated rats. In comparison with the radiated rats, those of the Liuweidihuang group exhibited nearly normal testicular structure, significantly lower MDA level (P 〈0.05) , bax expression (P〈0.01) , and bcl-2 expression (P 〈0.01). Conclusion: Liuweidihuang Pills can improve cellphone electromagnetic radiation-induced histomorphological abnormality of the testis tissue and re- duce its oxidative damage and cell apoptosis.
