不同物种成熟精子Fas蛋白的表达研究

摘要 使用免疫荧光标记和激光共聚焦显微镜方法,对不同物种成熟精子Fas蛋白进行观测。明确细胞凋亡蛋白Fas在不同物种成熟精子表面的分布规律,并对Fas蛋白进行定位。结果显示Cy3红色荧光标记的Fas蛋白主要分布在啮齿类动物(大鼠、小鼠),反刍动物(山羊、绵羊、牛)的精子头部和中段线粒体鞘上,而在猪和人的成熟精子上未检测到Fas蛋白。说明使用激光共聚焦显微镜技术可以精确定位标记后的Fas蛋白,不同物种成熟精子Fas蛋白的表达存在差异。 Fas protein in mature sperm of different species were observed by labeling and laser confocal microscopy. The objective of this study is to confirm the distribution of cell apoptosis protein Fas in mature sperm of different species and the localization of Fas protein. The results showed that Fas protein labeled red fluorescent Cy3 mainly distributed in sperm head and mitochondria sheath in midpiece in the rodent animal (rat and mouse), ruminant (goat, sheep and bull), but not in mature sperm of human and boar. This suggested that using confocal laser scanning microscopy could locate the labeled Fas protein in different species, accurately, and the expression of Fas protein is different in mature sperm of different species.
