迷迭香酸对单侧输尿管梗阻大鼠TGF-β_1、结缔组织生长因子表达的影响 被引量:1

摘要 目的观察迷迭香酸(RA)对单侧输尿管梗阻(UUO)大鼠肾间质纤维化中TGF-β1、结缔组织生长因子(CTGF)表达的影响。方法结扎大鼠左侧输尿管制作UUO模型,以RA为干预因素,应用免疫组化半定量分析、ELISA法检测肾组织TGF-β1、CTGF蛋白的表达。结果 UUO组、UUO+RA组肾组织TGF-β1、CTGF蛋白的表达水平均明显高于假手术(SOR)组(均P<0.05);与UUO组比较,UUO+RA组TGF-β1、CTGF的表达水平均明显降低(均P<0.05)。结论迷迭香酸具有一定的抗肾间质纤维化作用,其机制可能与抑制TGF-β1、CTGF的表达有关。 Objective To observe the effect of rosmarinic acid (RA) on expression oi transiormmg growth tactor-β1 (TGF-β1) and connective tissue growth factor (CTGF) in the model of rat kidney with unilateral ureteral obstruction (UUO). Methods UUO model was established by ligating the left ureter of the rats, rosmarinic acid was regarded as the interference factors. The protein expression of TGF-β1 and CTGF were detected by immunohistochemistry and ELISA. The immunohisto- chemistry was analyzed semi-quantitative by IPP 6.0. Results Compared with that of group SOR, the expression level of TGF-β1 and CTGF in renal tissue of group UUO and group UUO+RA was significantly increased (P〈0.05). Compared with group UUO, the expression level of TGF-β1 and CTGF in renal tissue of group UUO+RA was significantly reduced (P〈0.05). Con- clusion Rosmarinic acid has certain resistance to renal interstitial fibrosis, and its mechanism may be related to inhibit the ex- pression of TGF-β1 and CTGF.
