摘要
目的探讨芩丹颗粒对微波辐射小鼠心脏损伤的防护作用。方法 60只小鼠随机分为正常对照组、100 m W·cm-2辐射损伤组、辐射损伤+芩丹颗粒2.5,5.0和10.0 g·kg-1防护组。芩丹颗粒防护组小鼠于辐射前7 d每天ig给药1次。辐射后6 h,取静脉血,用生化法检测血清谷草转氨酶(GOT)、乳酸脱氢酶(LDH)和肌酸激酶(CK)活性。取心肌组织,用比色法检测三磷酸腺苷(ATP)含量、线粒体呼吸链复合体Ⅳ和Ⅴ活性,用光镜和电镜观察心肌组织结构的变化,用Western蛋白印迹法检测细胞色素c(Cyt c)和激活型胱天蛋白酶3表达的变化。结果与正常对照组比较,微波辐射组小鼠心肌细胞水肿、线粒体数量减少且形态异常,血清中GOT,CK和LDH活性升高(P<0.01),线粒体呼吸链复合体Ⅳ和Ⅴ活性显著下降(P<0.01),心肌细胞Cyt c和激活型胱天蛋白酶3蛋白表达显著升高(P<0.01)。与辐射损伤组比较,芩丹颗粒5.0和10.0 g·kg-1防护组小鼠心肌细胞体积正常,细胞核和细胞器形态完好,心肌细胞结构损伤程度显著缓解;心肌组织ATP含量、线粒体呼吸链复合体Ⅳ和Ⅴ活性均显著升高(P<0.01);心肌细胞Cyt c和激活型胱天蛋白酶3蛋白表达均显著下降(P<0.01)。结论芩丹颗粒对微波辐射致小鼠心肌细胞损伤具有一定的防护作用,其机制可能与其改善微波所致心肌细胞能量代谢有关。
OBJECTIVE To investigate the protective effect of Qindan granules on the heart of mice irradiated by microwave. METHODS Sixty mice were randomly divided into 5 groups: normal control group, 100 mW. cm-2 microwave radiation injury group, and Qindan granules 2.5, 5 and 10 g. kg-1 protective groups. Six hours after microwave irradiation, Qindan granules were ig given to the mice of Qindan granules protective groups once daily for 7 d. After 6 h of treatment, blood was collected. The activities of glutamic oxalacetic transaminase (GOT), lactate dehydrogenase (LDH) and creatine kinase (CK) in serum were detected by the biochemical method. The adenosine triphosphate (ATP) content and the activity of mitochondrial complexes IV and V in heart tissue were measured by colori- metric method. The changes in tissue structure were observed by light microscopy and electron micros- copy. The expression of cytochrome c (Cyt c) and cleaved caspase 3 was detected by Western blotting. RESULTS Myocardial cells developed edema. The number of mitochondria decreased while abnormal morphology, the content of GOT, CK and LDH increased in serum (P〈0.01). The activity of ATP and mitochondrial complexes IV and V decreased significantly (P〈0.01) compared with normal control in cardiac tissue and myocardial cells were also injured. The expression of Cyt c and cleaved caspase 3 was increased in myocardial cells (P〈0.01). Compared with microwave radiation group, the activity of ATP and mitochondrial complexes IV and V increased significantly in Qindan granules 2.5, 5 and 10 g·kg^-1 protective groups (P〈0.01). The injured myocardial cells were improved and the structure of nuclei and organelles was normalized. The expression of Cyt c and cleaved caspase 3 was decreased (P〈0.01). CONCLUSION Qindan granules has protective effect on microwave radiation induced injury in the mouse heart. The mechanism may be related to the improvement of energy metabolism of myocardium injured by microwave.
