摘要
目的探讨GLI1和MMP9在胰腺癌中的表达及其临床意义。方法使用免疫组织化学生物素蛋白-过氧化物酶(SP)法检测48例原发性胰腺癌和11例正常胰腺组织中GLI1和MMP9的表达。结果 1)在胰腺癌组织中,GLI1的高表达率和MMP9的阳性表达率分别为66.7%和62.5%,明显高于正常组织(P<0.05)。GLI1的高表达和MMP9的阳性表达与胰腺癌的浸润和转移有关(P<0.05)。2)GLI1高表达者生存时间明显低于低表达者(Log-rank检验,Χ2=12.230,P=0.000);MMP9阳性表达者生存时间明显低于阴性表达者(Log-rank检验,Χ2=4.077,P=0.043)。3)GLI1和MMP9在胰腺癌组织中表达呈正相关(r=0.548,P=0.000)。结论 GLI1可通过调控MMP9的表达促进胰腺癌细胞的侵袭和转移,两者过表达提示预后不良;对GLI1、MMP9的检测可有效地评估胰腺癌患者的预后。
Objective To study the expression and the clinical pathological significances of glioma-associated oncogene homolog-1(GLI1) and matrix metalloproteinases-9(MMP9)in pancreatic carcinoma. Methods The expression of GLI1 and MMP9 were detected by immunohistochemical method( S-P) in 48 cases of pancreatic carcinoma and 11 cases of normal pancreatic tissue. Results 1. The overexpression rate of GLI1 and the positive expression rate of MMP9 were 66. 7% and 62. 5%, and were remarkably higher in the tissue of pancreatic carcinoma than those in normal pancreatic tissue ( P〈0. 05). Overexpression of GLI1 and positive expression of MMP9 were positively correlated to the invasion and metas-tasis of pancreatic carcinoma(P〈0. 05). 2. The survival time of pancreatic carcinoma patients with the overexpression of GLI1 was shorter than the low expression(Log-rank test,χ2 =12. 230,P=0. 000);The survival time of pancreatic carcino-ma patients with the positive expression of MMP9 was shorter than the negative expression( Log-rank test,χ2 =4. 077,P=0. 043);The expression of GLI1 was positively correlated to that of MMP9 in pancreatic carcinoma. ( r = 0. 548, P=0. 000). Conclusion GLI1 may promote the invasion and metastasis of pancreatic carcinoma through upregulating the expression of MMP9. The overexpressions of GLI1 and MMP9 suggest the worse prognosis in pancreatic carcinoma. The ex-amination of GLI1 and MMP9 is useful in evaluating the prognosis of pancreatic carcinoma patients.
