摘要
目的探究细胞培养液中糖浓度对人牙周膜成纤维细胞原代培养成功率的影响。方法取60颗健康牙,随机分成3组,每组20颗,分别采用低糖、高糖、低糖+甘露醇培养液对其牙周膜成纤维细胞进行原代培养。记录每组细胞原代培养成功数,以及首次传代时间、传代后获得稳定的成纤维细胞系的成功数。结果人牙周膜成纤维细胞原代培养的成功率依次为低糖:90.00%,高糖:60.00%,低糖+甘露醇:85.00%。结论低糖较高糖培养液更有利于提高人牙周膜成纤维细胞原代培养的成功率。
Objective To Explore whether the glucose concentration of cell culture medium can affect the success rate of primary culture of human periodontal ligament fibroblasts or not. Methods There are three groups which were used low glucose medium、high glucose medium and low glucose added mannitol medium,and ten teeth was cultured every group. Then, compare the success rate of primary culture,the first time of transfer of culture, and the success rate of passage cells. Morphology, immuneocytochemistry were used to identify the cell source. Results The success rate in primary culture was,low glucose 90%,high glucose 60%,low glucose added mannitol 85%. Conclusion Compared with high glucose medium, low glucose medium will increase the success rate of primary culture of human periodontal ligament cells.
