摘要
目的观察小柴胡汤对胶原诱导性关节炎(CIA)大鼠的抗炎作用,并探讨其可能的作用机制。方法以胶原诱导性关节炎(CIA)大鼠为动物模型,随机分为正常对照组、CIA模型组、雷公藤多苷对照组、小柴胡汤高剂量组和小柴汤低剂量组,评定关节炎指数,光镜观察关节滑膜病理学变化,ELISA法测定不同时间点各组大鼠血清中TNF-α、IL-6、IL-10的水平。结果小柴胡汤高、低剂量组均可使大鼠踝关节炎指数明显减少,与模型组比较差异显著(P<0.05)。光镜下观察,滑膜组织水肿、滑膜细胞增生及炎性细胞浸润明显改善。ELISA结果显示,小柴胡汤高、低剂量组均可降低TNF-α、IL-6浓度,升高IL-10浓度,与模型组比较差异显著(P<0.01),其中小柴胡汤高剂量组作用尤为显著。结论小柴胡汤对CIA大鼠具有较强的抗炎作用,其作用机制可能与TNF-α、IL-6、IL-10等炎症介质的表达有关。
The present study was designed to investigate the anti-inflammatory effect of Xiaochaihutang in rats with collagen-induced arthritis (CIA) and the possible mechanisms. Rheumatoid arthritis model was constructed in rats by type II collage induction. The rats were divided into normal group, model group, tripterygium glycosides group, high and low doses of Xiaochaihutang groups. The anti-inflammatory effect of Xiaochaihutang was evaluated by assessing arthritis index, observing the pathological changes of rat feet joint synovial tissues by light microscope, and detecting the levels of TNF-α, IL-6, IL-10 by ELISA at various time points. Data showed that compare with model group, the arthritis index of high and low doses of Xiaochaihutang groups reduced significantly (P〈 0.05), and the synovial tissue edema, synovial cell hyperplasia and inflammatory cell infiltration were observed reduced; compared with model group, the high and low doses of Xiaochaihutang reduced the levels of TNF-α, IL-6, increased the levels of IL-10 (P〈 0.01), and the effect of high does Xiaochaihutang group was most obvious. These results indicated that Xiaochaihutang possesses a potent anti-inflammatory effect in CIA rats, and the possible mechanism may relate to the expressions of TNF-α, IL-6 and IL-10.
