电针对帕金森病大鼠中脑黑质26S蛋白酶体及核转录因子κB的影响 被引量:15

Effect of Electroacupuncture on 26 S Proteasome and Nuclear Factor kappa B in Substantia Nigra of Rats with Rotenone-induced Parkinson's Disease

摘要 目的:探讨26S蛋白酶体、核转录因子κB(NFκB)在电针对鱼藤酮致帕金森病(Parkinson’s disease,PD)模型大鼠保护机制中的作用。方法:将48只健康雄性SD大鼠随机分为正常组、假手术组、模型组、电针组,每组12只。模型组和电针组采用颈背部皮下注射鱼藤酮法造模40d。造模成功后电针组取"风府""太冲"行电针治疗(连续波,2Hz,1mA,20min/次,1次/d,连续28d)。观察各组大鼠行为学改变并评分,应用免疫组化法观察大鼠中脑黑质酪氨酸羟化酶(tyrosine hydroxylase,TH)的表达,免疫蛋白印迹法检测黑质TH、26S蛋白酶体、NFκB表达。结果:与正常组及假手术组比较,模型组大鼠出现明显的行为学改变,电针组行为学评分较模型组显著降低(P<0.05);与正常组及假手术组比较,模型组大鼠中脑黑质TH阳性神经元细胞计数、含量明显降低,电针组TH阳性细胞数、含量较模型组明显升高(均P<0.05);26S蛋白酶体在模型组表达较正常组及假手术组明显下降,电针组与模型组比较明显升高(均P<0.05);NFκB在模型组表达较正常组及假手术组明显增高,电针组与模型组比较明显降低(均P<0.05)。结论:电针显著改善鱼藤酮所致PD模型大鼠的行为学表现和评分,显著改善鱼藤酮所致黑质多巴胺能神经元丢失,从而发挥防治PD作用,其可能机制与增强蛋白酶体系统的功能有关,与其降低NFκB表达有关。 Objective To explore the role of 26 Sproteasome and nuclear factor kappa B(NFκB)in substantia nigra in the management of Parkinson's disease(PD)by acupuncture.Methods Forty-eight male SD rats were randomly divided into normal,sham operation,model and electroacupuncture(EA)groups(12rats/group).The PD model was established by 40-day consecutive subcutaneous injection of rotenone(1mg/kg dissolved in dimethyl sulfoxide and normal saline)at the back shoulder.The rats in the sham operation group were treated by subcutaneous injection of dose of saline."Fengfu"(GV 16)and"Taichong"(LR 3)acupoints were stimulated with EA at 2Hz,1mA,20 min of duration in each treatment,and daily for 28 consecutive days.The behavioral changes of rats in each group were measured and scored at 40 th day and 68 th day,respectively.Immunohistochemistry was used to determine the expression of tyrosine hydroxylase(TH)used to detect the expression of 26 Sproteasome and NFκB and TH were measured by Western blot.Results In comparison with the normal and sham operation groups,the behavioral scores of rats in the model group were elevated,which were significantly decreased by EA intervention(P〈0.05).The expression of TH and 26 Sproteasome decreased whereas the NFκB increased in the rats of model group(P〈0.05),and EA intervention reversed these changes(all P〈0.05).Conclusion EA intervention can improve PD rats' behavioral changes,which is pobably related to its effects in rcducing loss of TH-positive neurons,down-regulating NFκB protein expression,and upregulating 26 Sproteasome protein expression in the substantia nigra.
