半夏泻心汤对糖尿病胃轻瘫大鼠胰岛素抵抗的影响 被引量:50

Impact of Banxia Xiexin Tang( 半夏泻心汤) on Insulin Resistance in Diabetic Gastric Paralysis Rats

摘要 目的探讨半夏泻心汤对糖尿病胃轻瘫大鼠胰岛素抵抗的影响及可能作用机制。方法将120只大鼠随机分为空白组10只和造模组110只。造模组建立糖尿病胃轻瘫大鼠模型,造模成功的60只大鼠随机分为模型组、二甲双胍组[0.3 g/(kg·d)]、吗叮啉组[0.005 g/(kg·d)]及半夏泻心汤高[1.7 g/(kg·d)]、中[0.85 g/kg)、低[0.425 g/(kg·d)]剂量组各10只。连续灌胃1周,测定各组大鼠胃残留率、肠推进率,胃窦组织胃动素(MOT)、血管活性肽(VIP)、生成抑素(SS)和P物质(SP)含量,空腹血糖(FGB)、血清胰岛素(FINS)及胰岛素抵抗指数(IRI)。结果与空白组比较,模型组大鼠胃残留率及肠推进率,胃窦组织VIP、SS、MOT、SP水平,FGB、IRI及FINS差异均有统计学意义(P<0.05或P<0.01)。与模型组比较,半夏泻心汤高、中剂量组胃残留率和胃窦组织VIP水平明显下降;半夏泻心汤高、中、低剂量组胃窦组织SS水平和给药前后FGB水平、IRI明显下降,胃窦组织MOT、SP水平明显升高(P<0.05或P<0.01);半夏泻心汤中剂量组肠推进率明显升高(P<0.05)。结论半夏泻心汤能缓解糖尿病胃轻瘫大鼠症状,促进胃肠吸收功能,降低血糖,从而改善胰岛素抵抗。 Objective To discuss the impact of Banxia Xiexin Tang on insulin resistance in diabetic gastric paralysis rats and the possible mechanism. Methods Totally 120 rats were randomly divided into blank group( n =10) and DM modeled group( n = 110). In DM model group,DM models were established by intraperitoneal injection of streptozotocin( 40 mg / kg). Sixty successfully modeled rats were randomly divided into model group,metformin group( 0. 3 g / kg),domperidone group( 0. 005 g / kg),Banxia Xiexin Tang high dose group( 1. 7 g / kg),middle dose group( 0. 85 g / kg) and low dose group( 0. 425 g / kg),10 rats in each group,by gavage continuously for 1week. Detected gastric residual rate,alvine pushing rate,content of motilin( MOT),vasoactive peptide( VIP),somatostatin( SS) and substance P( SP) in gastric antrum,fasting blood glucose( FGB),serum insulin( FINS) and insulin resistance index( IRI). Results Comparing with blank group,gastric residual rate,alvine pushing rate,content of VIP,SS,MOT and SP in gastric antrum,FGB,IRI and FINS of rats in model group had significant difference( P〈0. 05 or P〈0. 01). Comparing with model group,gastric residual rate and content of VIP in gastric antrum in Banxia Xiexin Tang high and middle dose groups decreased obviously; SS in gastric antrum,FGB and IRI in Banxia Xiexin Tang high,middle and low dose groups decreased obviously while content of MOT and SP in gastric antrum increased obviously( P〈0. 05 or P〈0. 01); alvine pushing rate in Banxia Xiexin Tang middle dose group increased significantly( P〈0. 05). Conclusion Banxia Xiexin Tang could promote the recovery of gastrointestinal motility of DM rats,which may related to decreasing blood glucose,improving insulin resistance and gastrointestinal hormones.
