miRNA-126与颈动脉粥样硬化患者斑块稳定性的关系 被引量:5

摘要 目的研究颈动脉粥样硬化患者中外周血小分子RNA-126(miR-126)与斑块稳定性之间的关系。方法选择可能存在颈动脉硬化的患者,根据颈动脉超声检测结果分为正常组、稳定斑块组和不稳定斑块组,常规检测血压、体重指数、肝肾功能、血糖、血脂、尿酸、超敏C反应蛋白(hs CRP),real time PCR检测外周血中miRNA-146a的表达水平。对存在颈动脉粥样硬化的患者予瑞舒伐他汀20 mg每晚一次口服治疗,12个月后复查颈动脉超声、hs CRP及miR-126。结果 3组患者的年龄、体重指数、血压、血糖、血脂、尿酸、肝肾功能均无明显差异(P>0.05),不稳定斑块组患者的hs CRP明显升高(P<0.05)。与正常组相比,稳定斑块组患者血浆中的miR-146a水平上升(P<0.05),不稳定斑块组患者的miR-126水平更加显著(P<0.01)。治疗12个月后,16例不稳定型动脉粥样硬化患者的颈动脉斑块稳定性改善,同时外周血hs CRP及miR-126也明显下降(P<0.05)。结论 miR-126可能与颈动脉粥样硬化发展的发生发展相关。 Objective To investigate the association between miRNA-126 and carotid atherosclerosis. Methods106 cases of patients enrolled in our hospital were divided into normal group、stable plaque group and unstable plaque group according to carotid ultrasonography,whose blood pressure、body mass index( BMI) 、liver function( GPT) 、renal function( serum creatinine) 、fasting blood glucose、blood fat、uric acid and hyper sensitive C-reactive protein( hs CRP) were routinely measured. The circulatory level of miRNA-146 a were explored by real time polymerase chain reaction technology. 16 patients with unstable carotid plaque were recalled for the measurements of carotid ultrasonography、blood hs CRP and miR-126 after therapy for 12 months by Rosuvastatin 20 mg qn. Results There were not significant differences in age、blood pressure、BMI、GPT、serum creatinine、blood glucose、blood fat( P〈0. 05). Compared with normal patients,the level of hs CRP in unstable plaque patients significantly increased( P〈0. 05). The circulatory level of miRNA-146 a in stable plaque patients was higher than that in normal patients( P〈0. 05),however,this rise was more significant in unstable plaque patients( P〈0. 01). The plaque stability was improved after therapy by Rosuvastatin accompanied by the decreased level of circulatory hs CRP and miR-126( P〈0. 05). Conclusion miRNA-146 a may be related with the pathological progress of carotid atherosclerosis.
