摘要
目的观察利多卡因对自发性及催产素诱发人子宫平滑肌收缩的影响并探讨其相关机制。方法子宫平滑肌取自择期行剖宫产术患者子宫。使用体外肌肉等长张力测定试验测定不同浓度利多卡因(10-5,5×10-5,10-4,5×10-4 mol/L)对自发性及催产素诱发人子宫平滑肌收缩的影响。使用钙离子荧光指示剂Fluo-3孵育子宫平滑肌以检测细胞内钙离子浓度。使用Western blot法检测子宫平滑肌催产素受体及磷酸化PKC表达水平。结果与对照组比较,10-4及5×10-4 mol/L利多卡因可导致自发性收缩和催产素处理的子宫平滑肌收缩幅度和收缩频率减少(均P<0.05)。同时,10-4及5×10-4 mol/L利多卡因可导致自发性收缩和催产素处理的子宫平滑肌细胞内钙离子浓度减少(均P<0.05)。Western blot检测结果显示,10-4及5×10-4 mol/L利多卡因可导致自发性收缩和催产素处理的子宫平滑肌细胞内磷酸化PKC表达水平降低(均P<0.05)。结论利多卡因可通过降低细胞内钙离子浓度导致自发性及催产素诱发人子宫平滑肌收缩功能减弱。
Objective To investigate effects of lidocaine on spontaneous and oxytocin-induced contractions of human myometrial smooth muscles and its possible mechanisms.Methods Myometrial strips were obtained from women undergoing selective prelabour caesarean section.The effects of different concentrations of lidocaine(10-5,5×10-5,10-4,5×10-4 mol/L)on spontaneous and oxytocin-induced contractions of human myometrial smooth muscles were examined with a strain gauge transducer.These strips were also loaded with the calcium indicator Fluo-3for assessment of intracellular calcium concentrations.Western blot was applied to detect the protein levels of oxytoxin receptors and PKC phosphorylation.Results As compared with control group,10^-4 and 5×10^-4 mol/L of lidocaine induced a significant decline in contractile amplitude and frequency of non-oxytoxin and oxytoxin-treated myometrial smooth muscles(P〈0.05).Meanwhile,10^-4 and 5×10^-4 mol/L of lidocaine also significantly decreased intracellular Ca2+concentration in non-oxytoxin and oxytoxin-treated myometrial smooth muscles(P〈0.05).Incidentally,we also found that 10-4 and 5×10^-4 mol/L of lidocaine leaded to a significant decrease in protein levels of PKC phosphorylation in non-oxytoxin and oxytoxin-treated myometrial smooth muscles(P〈0.05).Conclusion Lidocaine attenuates spontaneous and oxytocin-induced contractions of human myometrial smooth muscles by down-regulating the intracellular Ca2+concentration.
