摘要
为了研究扇贝糖蛋白(AIG)的抗肿瘤活性,选用昆明种小鼠为试验动物,通过移植性肿瘤动物模型,研究不同剂量的AIG(80,160,320 mg/kg bw)对肉瘤-180(S180)生长的抑制作用。设3个剂量组,1个溶剂对照组和1个模型对照组。结果表明,AIG在80,160,320 mg/kg bw 3个剂量水平对S180的生长均有抑制作用,与模型对照组相比均存在极显著差异(P<0.01),呈剂量-反应关系。低、中、高剂量组的抑瘤率分别为31.83%,34.50%和36.36%;不同剂量的AIG均可抑制S180荷瘤小鼠脾脏淋巴细胞的凋亡,且早期凋亡率及总凋亡率与AIG的剂量正相关。当剂量为320 mg/kg bw时,AIG抑制脾脏淋巴细胞凋亡效果最佳,与模型对照组相比存在显著差异(P<0.05);与模型对照组相比,不同剂量的AIG均可上调荷瘤小鼠外周血CD3+、CD4+的百分比及CD4+/CD8+的比例,且与AIG的剂量正相关。对CD8+的影响无剂量-反应关系。与模型对照组相比,中剂量组、高剂量组CD3+、CD4+及CD4+/CD8+差异显著(P<0.05,P<0.01)。结果提示:AIG具有较好的抗肿瘤活性,该活性可能与其抑制荷瘤鼠脾脏淋巴细胞凋亡,上调其外周血T淋巴细胞亚群中CD3+、CD4+的百分比及CD4+/CD8+比值有关。
To investigate the anti-tumor activity and the influence on immunologic function in mice of glycoprotein from Argopectens irradias (AIG), the research selected Kunming mice as experimental animals. S180 tomor-bearing mice were used as animal model. The experiment setted three AIG dose groups (80, 160, 320 mg/kg bw), a solvent control group and a model control group. The tumor growth were measured, the effects on spleen cell apoptosis and T-Lympho- cyte Subsets were determined by flow cytometry. The results showed that tumor weights were decreased significantly in a dose dependent manner in AIG treated groups compared with the control group (P〈0.01). The inhibitory rates were 31.83%, 34.50% and 36.36% in the low-dose, middle-dose and high-close AIG groups respectively. Compared with the control group, all doses of AIG can inhibit the apoptosis of splenic lymphocytes of S180 tumor-burdened mice. Early apoptosis rate and total apoptosis rate had a positive correlation with the dose of AIG, which means the high-dose (320 mg/kg bw) is the most effective one (P〈0.05). In addition, the proportion of the CD8+ T cell was not affected, the per- centage of CD3+ T cells and CD4+ T cells were significantly increased in middle-dose and high-dose groups that com- pared with control group (P〈0.05, P〈0.01). Furthermore, there was a definite trend toward an increased ratio of CD3+T cell, CD4+ T cell and CD4+/CD8+ in the AIG administered groups in dose-dependent manner. This increase of the ratio may be achieved the best effect when the mice were administered With high-dose of AIG. The results indicate that AIG had some anti-tumor effect between 80N320 mg/kg bw, this kind of activity may result from restraining the apoptosis of spleen cells in S180 tumor-bearing mice and upregulating the percentage of CD3+ T cell, CD4+ T cell and CD4+/CD8+ in S180 tumor-bearing mice peripheral blood.
