高效液相色谱法分析龙眼多糖的单糖组成 被引量:4

摘要 以新鲜龙眼果肉为原料,采用热水浸提法提取多糖,其提取工艺条件为:料液比(W∕V)1∶3,提取温度90℃,提取时间2 h.2次重复实验的龙眼多糖得率为0.52%,总糖含量为20.23%,还原糖含量3.45%.多糖经硫酸水解后,通过PMP柱前衍生化处理后,以V(水)∶V(甲醇)=50∶50作流动相,1.0 m L/min的流速,检测波长254 nm的色谱条件,利用高效液相色谱法进行分析.结果表明,龙眼多糖中存在L-鼠李糖(L-Rha)、D-葡萄糖(D-Glc)、D-半乳糖(D-Gal)3种单糖. This research used the fresh Longan as material and hot water extracting method to extract polysaccha- rides. The extraction conditions included material - to - solution ratio 1 : 3, extraction temperature 90 ℃ extraction time 2 h and experiments were repeated two times. The content of Longan polysaccharide, total sugar and reducing sugar was 0. 52% ,20. 23% and 3.45% ,respectively. After the polysaccharides was hydrolyed by sulfuric acid ,the hydrolysate took PMP for Pre Column Derivatization, and the PMP derivatives were analysed by HPLC. With water: methanol(50: 50, V/V) as mobile phase, flow rate of 1.0 mL/min, at the wavelength of 254 nm analyse the PMP de- rivativesby HPLC analysis. The results show that Longan polysaccharide were composed of L - Rhamnose (L - Rha), D - Glucose ( D - Glc ), D - Galactose ( D - Gal) three kinds of monosaccharides.
