摘要
为了研究曼氏无针乌贼墨汁黑色素(melanin from Sepiella maindroni ink,MSMI)的抗衰老作用,将54只ICR小鼠随机分为6组,包括空白对照组,衰老模型组,阳性对照组(Vc组)和墨汁黑色素低[25 mg/(kg·d)]、中[120 mg/(kg·d)]和高[200 mg/(kg·d)]剂量组。除空白对照组皮下注射生理盐水外,其他组小鼠皮下注120 mg/(kg·d)的半乳糖。连续给药45 d后,分别测定小鼠血清、肝脏、脑、心脏中的丙二醛(MDA)含量、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性,测定肝脏指数,脾脏指数。结果显示与衰老模型组相比,曼氏无针乌贼墨汁黑色素(MSMI)能显著降低丙二醛(MDA)含量,黑色素组的CAT、SOD、GSH-Px活性及体重都显著提高。曼氏无针乌贼墨汁黑色素有一定的抗氧化、抗衰老能力。
In this study,the anti-aging effect of MsMI (melanin from Seplella maindroni ink) on mimetic aging mice in- duced by D-galactose was investigated. ICR mice was selected as animal model, which were randomly divided into six groups,including control group, D-galaetose group, positive control group ( Vc group), MSMI treated groups of low [ 25 mg/(kg · d) ] ,medium [ 120 mg/(kg v d) ] and high [200 mg/(kg· d) ] dosage. Except control greup,all the other groups were treated with 120 rag/( kg · d) D-galactose by subcutaneous injection. After a trial period of 45 days, the content of MDA (malondialdehyde) and the activities of CAT (catalase),SOD (superoxide dismutase) and GSH-Px (glutathione peroxidase) in serum, liver, brain and heart were measured. Besides, the liver and thymus index were meas- urod. The results showed that compared with D-galaetose group, MSMI can significantly decrease the contents of MDA and enhance the activities of CAT,SOD,GSH-Px in serum,liver,brain and heart of mice. Hence,it was concluded that MSMI can attenuate the increased oxidative damages in D-galactose-induced senescent mice and delay aging.
