HPLC法同时测定2种金银花复方制剂中的绿原酸和连翘苷的含量 被引量:8

摘要 目的采用HPLC法同时测定2种金银花复方制剂中绿原酸和连翘苷的含量。方法采用Phenomenex-C18(250mm×4.6mm,5μm);流动相:乙腈(A)1.0mL·L^-1磷酸水溶液(B)为流动相进行梯度洗脱;流速:1.0mL·min^-1;柱温:30℃;检测波长:双波长检测,绿原酸检测波长λ1为326nm,连翘苷检测波长λ2为228nm。结果绿原酸在0.010-0.100mg·mL^-1范围内线性关系良好,r=0.999 7,在2种制剂中的平均回收率均大于98.0%;连翘苷质量浓度在0.001-0.010mg·mL^-1范围的线性关系良好,r=0.999 8,在2种制剂中的平均回收率均大于97.0%。银翘解毒片中的绿原酸含量为0.546mg·片^-1,连翘苷含量为0.040mg·片^-1;复方金银花颗粒中绿原酸的含量为2.550mg·袋^-1,连翘苷为0.223mg·袋^-1。结论该方法灵敏度、精密度好,结果准确可靠。 Objective To determine the contents of chlorogenic acid and phillyrin in 2 honeysuckle compound preparations simultane‐ously by HPLC .Methods The samples were separated on a Phenomenex‐C18 (250 mm × 4 .6 mm ,5 μm) chromatographic column using acetonitrile‐1 .0 mL · L^ -1 phosphoric acid solution as a mobile phase with a gradient elution .The detection wavelength was at 326 nm (chlorogenic acid) or 228 nm (phillyrin) ,the flow rate was 1 .0 mL · min^-1 ,and the temperature of column was 30 ℃ . Results The linear range of chlorogenic acid was 0 .010‐0 .100 mg · mL^ -1 (r = 0 .999 7) ,and the average recoveries were more than 98 .0% ;the linear range of phillyrin was 0 .001‐0 .010 mg · mL ^-1 (r= 0 .999 8) ,and the average recoveries were more than 97 .0% .The contents of chlorogenic acid and phillyrin in Yinqiao Jiedu Tablets were 0 .546 and 0 .040 mg · tablet^-1 ,respectively ;The contents of chlorogenic acid and phillyrin in Compound Lonicera Granules were 2 .550 and 0 .223 mg · bag^-1 ,respectively .Conclu‐sion The method is sensitive ,accurate ,and can be used to control the quality of the 2 honeysuckle compound preparations .
