摘要
目的:观察健脾养正消癥方对BALB/c裸鼠人胃癌MGC-803细胞皮下移植瘤生长及转移的影响,并探讨其相关机制。方法:建立人胃癌MGC-803裸鼠皮下移植瘤模型,并随机分为阴性对照组、阳性对照组、健脾养正消癥方大、小剂量组,观察对裸鼠皮下移植瘤的抑制作用。使用荧光定量PCR检测移植瘤内RECK、c-myc、MMP-9、MMP-14、CXCR4和VEGFA的基因表达。Western blot检测移植瘤内的MMP-9、MMP-14、RECK、STAT3和p-STAT3的蛋白表达。结果:与阴性对照组比较,健脾养正消癥方大、小剂量组有明显缩小皮下移植瘤体积的作用。荧光定量PCR显示,健脾养正消癥方大剂量组中RECK的表达显著上调,而c-myc、MMP-9、MMP-14、CXCR4和VEGFA的表达均显著下调。Western blot检测显示,健脾养正消癥方大、小剂量组能上调RECK蛋白的表达,下调MMP-9、MMP-14的表达,并抑制p-STAT3表达。结论:健脾养正消癥方对胃癌MGC-803细胞裸鼠皮下移植瘤的生长有显著的抑制作用,其作用机制可能与该方调节胃癌组织中侵袭转移的相关基因、蛋白有关。
Objective: To observe the inhibitory rate of Jianpi Yangzheng Xiaozheng formla (JPYZXZ) on the transplanted tumor due to Gastric Cell Line MGC-803 in nude mice and to explore its molecular mechanism. Methods: Transplanted transplantation tumor nude model mice were made, and modeling nude mice were randomly divided into negative control group, positive control group and JPYZXZ formula group (high dose group, low dose group). The inhibitory rate of JPYZXZ formula on transplanted tumor was observed. Real-time PCR was used to detect the expression of RECK, C-MYC, MMP-9, MMP-14, CXCR4 and VEGFA from transplanted tumor. Western blot was used to detect the expression of MMP-9, MMP-14, RECK, stat3 and p-stat3. Results: The JPYZXZ formula high, low dose groups can inhibit the growth of transplanted tumor, with stastistic significant compared with negative control group. Comparing with the negative control group, gene expression of RECK upregulated and C-MYC, MMP-9, MMP-14, CXCR4, VEGFA downregulated in JPYZXZ formula group. Comparing with the negative control group, protein expression of RECK upregulated and MMP-9, MMP-14, p-stat3 in JPYZXZ formula group downregulated. Conclusion: JPYZXZ formula has significant inhibition effect on the transplanted transplantation tumor. Its mechanism may be related to invasion and metastasis related gene and protein in gastric cancer tissues.
