摘要
为了提高PHAMCL在门多萨假单胞菌NK-01中的积累,采用单因素实验和正交实验确立了发酵生产PHAMCL的最佳条件,即以PHA产量为指标的最佳发酵条件为15 g/L葡萄糖浓度、C/N=50、发酵时间48h,该条件下获得产量0.8 g/L以上的PHA;以PHA占菌体干重百分含量为指标的最佳发酵条件为10 g/L葡萄糖浓度、C/N=60、发酵时间48 h,该条件下获得占菌体干重50%以上的PHA。该研究将为门多萨假单胞菌NK-01用于PHAMCL的规模化生产提供理论依据。
In order to improve the accumulation of PHAMcL in vivo, single factor and orthogonal experiments were adopted to study the optimization fermentation conditions for Pseudomonas mendocina NK-01. When PHAMCL output was taken as an index, under optimized fermentation conditions at 15 g/L glucose, with C/N ratio at 50 and fermented for 48 h, produced over 0.8 g/L of PHAMcL. When the index was turned to PHAMcL percentage content of dried cell weight (DCW), the best fermentation conditions were 10 g/L glucose, with C/N ratio at 60 and fermented for 48 h, the PHAMcL output was over 50% of DCW. This study will provide theoretic foundations for large scale production of PHAMcL by P. mendocina NK-01.
