摘要
目的:探讨新疆哈萨克族高血压患者外周血单个核细胞激活与NLRP3炎症体通路的相关性。方法:随机选取2013-09-2014-02新疆医科大学第一附属医院高血压科门诊首次就诊且未经降压药物治疗的新疆哈萨克族高血压患者30例,年龄(54.2±5.7)岁,选取同期同民族健康体检者30例,年龄(53.7±5.4)岁,两组男女比例均为1∶1。采集两组受试者外周血并分别提取血浆和单个核细胞,采用ELISA法检测血浆白细胞介素(IL)-1β和IL-18含量;使用荧光定量RT-PCR检测单个核细胞中NLRP3、Caspase-1、IL-1β和IL-18mRNA相对表达水平;运用Western blot法检测单个核细胞中NLRP3和Caspase-1蛋白的相对表达量。结果:两组受试者基线资料差异无统计学意义(P>0.05);与健康对照组相比,高血压组血浆IL-1β和IL-18水平明显升高(P<0.05);外周血单个核细胞层NLRP3、Caspase-1、IL-1β和IL-18的mRNA相对表达量均高于对照组(P<0.05);NLRP3和Caspase-1蛋白的表达量也较对照组增高(P<0.05)。结论:新疆哈萨克族高血压患者外周血单个核细胞处于一定程度的炎症激活状态,且该炎症反应可能与NLRP3炎症体及其下游相关细胞因子的表达增多有关。
Objective:To explore the correlation of NLRP3 inflammasome pathway and inflammatory activation of peripheral blood mononuclear cells(PBMCs)in patients with essential hypertension(EH)in Xinjiang Kazak.Method:30untreated Kazakh hypertensive patients(mean age 54.2±5.7years)and 30 Kazakh healthy subjects(mean age 53.7±5.4years)were randomly collected between September 2013 and February 2014 in department of hypertension,the First Affiliated Hospital of Xinjiang Medical University.The male to female ratio was 1:1in each group.The plasma and PBMCs were separated,respectively.Enzyme linked immunosorbent assay(ELISA)was performed to measure the plasma levels of interleukin(IL)-1βand IL-18.The real time quantitative RT-PCR was used to detect the mRNA levels of NLRP3,Caspase-1,IL-1βand IL-18.Western blot analysis was further conducted to determine the protein levels of NLRP3 and Caspase-1.Result:No significance of general condition between the two groups was observed(P〉0.05).Results of ELISA revealed an elevated plasma level of IL-1βand IL-18 in patients with EH in Xinjiang Kazakh(P〈0.05).Compared to the healthy controls,the mRNA levels of NLRP3,Caspase-1,IL-1βand IL-18 were significantly increased in PBMCs of Kazakh hypertensive patients(P〈0.05);and the protein expressions of NLRP3 and Caspase-1were also up-regulated markedly(P〈0.05).Conclusion:The data suggest that PBMCs in patients with EH in Xinjiang Kazakh are inflammatory activated,and the increased expression of NLRP3 inflammasome and its downstream related molecules in plasma and PBMCs may be involved in the process of this inflammatory activation.
