摘要
目的:观察电针对创伤后应激障碍(PTSD)模型大鼠行为学的影响,并进一步研究电针对PTSD模型大鼠蓝斑核神经元型一氧化氮合酶(nNOS)表达的影响。方法:将雄性SD大鼠30只随机分为三组:正常对照组、模型组和电针组,每组10只。采用单程长时应激(SPS)方法建立PTSD模型,模型建立后电针大鼠"百会"穴和"足三里"穴,频率2Hz,强度3V,持续30min,连续1周。采用免疫组织化学方法观察nNOS在大鼠蓝斑的表达。结果:与正常对照组比较,模型组大鼠蓝斑nNOS阳性细胞数明显增加(P<0.01),平均灰度值显著降低(P<0.01);电针后大鼠蓝斑nNOS阳性细胞数明显减少(P<0.01),平均灰度值显著增加(P<0.01)。结论:电针可下调PTSD模型大鼠蓝斑nNOS的表达,推测这可能是电针治疗PTSD机制之一。
Objective :To observe the electrical behavior of rats to learn the impact of post‐traumatic stress disorder (PTSD) ,and further study the expression for the electric rat model of PTSD locus coeruleus neuronal nitric oxide syn‐thase (nNOS) impact .Methods :30 male Sprague‐Dawley rats were randomly divided into three groups :normal control group ,model group and electroacupuncture(EA) groups of 10 .Using one‐way long stress (SPS) method to establish PTSD model after model“Baihui”Point EA rats and“Zusanli”Point ,frequency 2Hz ,strength 3V ,sustained 30min ,for a week .Immunohistochemistry was used to observe the expression of nNOS in rat locus coeruleus .Results:Compared with the normal control group ,model group locus coeruleus nNOS positive cells was significantly increased (P〈0 .01) , significantly reduce the average gray value(P〈0 .01);rat locus coeruleus nNOS positive cells was significantly reduced after EA (P〈0 .01) ,a significant increase in the mean gray value (P〈0 .01) .Conclusion:EA can down regulate the expression of nNOS locus coeruleus PTSD rat model ,suggesting that this may be one mechanism of acupuncture treat‐ment of PTSD .
