摘要
目的:观察壮骨止痛方提取物对去势大鼠Dkk1蛋白的影响,并探究其治疗骨质疏松症的具体机制。方法:60只3月龄的健康雌性SD大鼠,随机分为壮骨止痛方组、模型组、假手术组、空白组,每组15只。壮骨止痛方组、模型组去势(去卵巢)后建立绝经后骨质疏松大鼠模型,假手术组去除卵巢周围相应体积脂肪。术后5d开始给药,13周后取血和骨组织,测定血清中雌激素(E2)、降钙素(CT)、骨钙素(BGP)含量变化、骨组织病理变化、骨小梁密度,以及骨组织中Dkk1、β-catenin蛋白的表达情况。结果:与模型组相比,壮骨止痛方能降低去势雌鼠血清BGP、CT含量,差异具有统计学意义(P<0.05);壮骨止痛方组E2与模型组相比,差异具有统计学意义(P<0.05)。壮骨止痛方组相比模型组,骨髓腔扩大不明显,且骨小梁形态较为正常;与模型组相比,壮骨止痛方组骨小梁密度有明显升高,具有显著性(P<0.05);与模型组比较,壮骨止痛方组骨组织中骨组织Dkk1蛋白的表达降低、而β-catenin蛋白的表达增多,差异具有统计学意义(P<0.01)。结论:壮骨止痛方可能是通过下调去势雌鼠体内Dkk1蛋白表达,进一步直接影响β-catenin的表达,间接影响雌激素、降钙素等激素的变化,从而达到改善骨代谢紊乱来治疗骨质疏松症。
Objective:To observe the effect of extracts from Zhuanggu Zhitong Prescreption on dickkopfl protein of ovariectomized female rats and explore the specific mechanism of it remedy osteoporosis. Methods : 60 three - month old healthy SD rats were divided into Zhuanggu Zhitong Prescription group, model group, sham - operated group and blank group, 15 rats in each group. After being ovariectomized, Zhuanggu Zhitong Prescription group and model group rats were used to establish post -menopause osteoporotic rats model. The sham -operated group rats cut the same volume fat surrounding the ovary. 5 day after operation, rats were given medicine. 13 weeks later,we drew blood and bone tissue and measured E2, CT, BGP level in serum, bone tissue pathological change, bone trabecula density and dickkopfl and β - catenin protein expression level. Results:Compared with the model group, Zhuanggu Zhitong Prescription made BGP and CT content in ovariectomized female rats serum decline. The discrepancy had statistical significance (P 〈 0.05). E2 from Zhuanggu Zhitong Prescription group compared with that of the model group had Statistical significance(P 〈0.05). Contrasted with model group,the Zhuanggu Zhitong Prescription groupg marrow cavity had no obvious enlarge and the form of bone trabecula was normal and the bone trabecula density increased a lot ( P 〈 0.05 ). Compared with the model group, Zhuanggu Zhitong Prescription groupg bone tissue dickkopfl protein expression was decreased, but β - catenin protein expression increased and the difference had statistical significance (P 〈 0. 01 ). Conclusion:Zhuanggu Zhitong Prescription may through the way that reducing the expression of dickkopfl protein in ovariectomized female rats and then influence the expression of β - catenin protein and regulate estrogen, CT, BGP content, indirectly to remedy osteoporosis.
