曲霉发酵啤酒糟培养基配方的优化

摘要 为更好地利用啤酒糟,从富含纤维素的土壤中分离和筛选纤维素高效降解的曲霉,在啤酒糟培养基中加入不同的N源、C源,并调整其比例,接种等量的活化曲霉孢子悬浮液,观察菌丝生长情况以及测定菌株的酶活力,筛选促进菌株生长的最优配方。结果表明,最适的碳源和氮源分别为乳糖和胰蛋白胨,其最佳添加量分别为6%和4%;促进曲霉生长的最优碳氮比为5︰7,发酵后维素酶活力达188.49 IU,葡萄糖含量为0.38 g。这表明啤酒糟可以作为曲霉的优良培养基,对选育优良的啤酒糟纤维素分解菌具有一定的指导意义。 In order to make better use of brewer's grain, Aspergillus which degraded cellulose effectively was separated and screened from cellulose-rich soil; and the mycelial growth was observed and the enzyme activity of strains was determined to screen the optimal formula which promoted the growth of the strains by adding different C sources and N sources at different proportions of them to brewer's grain medium inoculated with same amount of activated Aspergillus spore suspension in this study. The results showed that the optimal C source was lactose and the optimum additive rate was 6%; the optimal N source was tryptone and the optimum additive rate was 4%; and the optimal C and N ratio is 5:7 for the vigorous growth of Aspergillus, the CMC enzyme activity was up to 188.49 IU after fermentation, and the glucose content was 0.38 g. So the brewer's grain could be used as a good Aspergillus culture medium, which was of signifi cance for breeding fi ne brewer's grain cellulose decomposing microorganism or strains.
