摘要
目的:研究实验性大鼠胃溃疡期间海马区肠三叶因子(ITF)表达的变化。方法:选择SD大鼠42只,其中实验性胃溃疡36只,正常对照6只,用免疫组织化学染色和RT-PCR方法分别检测大鼠海马ITF蛋白表达量和mRNA转录变化。结果:大鼠海马锥体细胞和颗粒细胞质内,以及海马内部分神经细胞可见ITF免疫反应阳性物质。与正常大鼠比较,实验性大鼠溃疡1 d后海马ITF平均灰度值开始降低,随后逐渐降低,1周后达到最低水平(P〈0.01),维持2~4周后开始恢复(P〈0.05)。RT-PCR检测表明,大鼠实验性溃疡后1 d开始,海马ITF mRNA转录水平逐渐升高,1周达到最高值,其后开始下降(P〈0.05~0.01),4周后接近实验前水平。结论:实验性胃溃疡大鼠海马中ITF高表达可能是溃疡愈合过程神经内分泌调节的重要因素。
Objective: To explore the expressions of intestinal trefoil factor (ITF) in hippocampus in experimental rat with gastric ulcer. Methods:Fouty-two SD rats were selected, thirty-six rats were used as experimental gastric ulcer and 6 rats as normal controls. The changes of ITF protein expressions and mRNA transcription in hippocampus of experimental gastric ulcer and normal rats were tested by immunohistochemistry and RT-PCR. Results:ITF immunoreaetive substances were identified in cytoplasm of hippocampal pyramidal cells and granular cells, and part of the nerve cells. Compared with normal rats, average gray value of hippocampal for ITF immunoreactivity began to decrease in experimental ulcerative rats at 1 d, then gradually decreased and reached the lowest level at 1 week (P 〈0.01 ) and started to recover (P 〈 0.05 ) after maintaining 2 -4 weeks. RT-PCR detection showed that in the rats with gastric ulcer, hippocampal ITF mR- NA transcription level increased gradually at 1 d post ulcer and reached the highest value at 1 week, then began to decline (P 〈 0.05 -0.01 ), and four weeks later the ITF mRNA approximated the level before the experiment. Conclusion:The results show that the high expression of ITF in hippocampus may be an important factor in the regulation of neuroendocrine healing process for experimental gastric ulcer in rats.
