摘要
目的:采用HPLC法以芦丁、木樨草素、槲皮素、山柰酚、异鼠李素、白杨素为指标对蒺藜进行含量测定方法研究,并对比不同产地6种黄酮类成分的区别。方法:采用HPLC法,色谱柱为Welch Ultimate LP-C18(250 mm×4.6 mm,5μm),流动相为0.05%磷酸水-乙腈梯度洗脱,检测波长为360 nm,流速为1.0 ml·min-1,对蒺藜6种黄酮成分进行含量测定。结果:对照品芦丁、木樨草素、槲皮素、山柰酚、异鼠李素、白杨素的标准曲线呈良好的线性关系(r≥0.999 4),平均回收率为97.5%~99.7%,RSD≤1.24%(n=6),符合方法学的相关要求。各产地6种成分的含量各有不同,但从总量上来看,以湖南产的黄酮总量最高为15.59 mg·g-1,以贵州产的黄酮总量最低为7.89 mg·g-1。结论:所建立的HLPC法,简便易行,精密度、回收率、重复性等均符合要求,可用于蒺藜的含量测定,6种蒺藜黄酮成分在不同产地蒺藜药材普遍存在,以此为目标进行相关研究具有意义。
Objective:To establish an HPLC method for the determination of rutin, luteolin, quercetin, kaempferol, isorhamnetin and chrysin in Tribulus terrestris L. Methods:The chromatographic column was Welch Ultimate LP-C18(250 mm ×4.6 mm, 5 μm), the mobile phase was acetonitrile and 0. 05% phosphoric acid with gradient elution,the detection wavelength was 360 nm,and the flow rate was 1. 0 ml·min-1 . Results: The linear relationship of standard curve of rutin, luteolin, quercetin, kaempferol, isorhamnetin and chrysin was good (r≥0. 999 4). The average recovery was 97. 5%-99. 7% with RSD≤1. 24% (n=6). Conclusion:The meth-od is simple, and the precision,recovery and repeatability all meet the requirements,which can be used for the content determination of Tribulus terrestris L.
