摘要 本研究以"红衣少女"香石竹组培苗为材料,通过根尖压片实验对"红衣少女"香石竹染色体计数方法进行探索,确定"红衣少女"香石竹染色体制片的最佳技术参数,从而对其倍性进行鉴定。本实验结果表明:以早上8:30用8-羟基喹啉处理1.5h为最佳,酸解时间以6-8min为宜。染色体压片结果显示:"红衣少女"香石竹染色体数为27条,鉴定其为3倍体。 In this paper, Dianthus caryophyllus L. cv Hongyishaonv plant chromosome number was researched by the method of root- tip cell chromosome pressing technology with the breeding Dianthus caryophyllus L, cv Hongyishaonv as the test materials. The objective of this research was to confirmed the ploidy of Dianthus caryophyllus L. cv Hongyishaonv and the best technical parameters. The result showed that 1.5h was the best time with soaking of 8-hydroxyquinoline at 8:30 Am. The acidolysis time of root-tip should be controlled within 6-8 min. The chromosome number of Dianthus caryophyllus L. cv Hongyishaonv was 27(2n=3x =27).
