摘要
目的观察不同剂量的野百合碱诱导大鼠肺动脉高压模型的建立,探索合适的野百合碱造模剂量。方法将60只雄性SD大鼠随机分成4组,正常对照组、野百合碱低剂量组、野百合碱中剂量组和野百合碱高剂量组,一次性给予颈背部皮下注射不同剂量野百合碱诱导建立肺动脉高压大鼠模型。4周后,测量各组大鼠肺动脉压力、心输出量、肺血管阻力以及右心室肥厚程度。结果野百合碱高剂量组大鼠死亡率、肺动脉压力、右心室室壁厚度高于中剂量组、低剂量组和正常对照组(P<0.001),心输出量低于中剂量组、低剂量组和正常对照组(P<0.001);高剂量组肺动脉压力、肺血管阻力高于中剂量组、低剂量组(P<0.001)。结论中剂量组野百合碱诱导的大鼠肺动脉高压模型更为接近大鼠肺动脉模型的理想数值。
Objective To investigate the different doses of monocrotaline induced pulmonary hypertension and explore situable monocrotaline dose to model pulmonary hypertension.Methods Sixty male Sprague Dawley rats were randomly divided into four groups, normal control group, low dose group, middle close group and high dose group, rats were given subcutaneous injection of appropriate doses of monocrotaline disposably in order to induce the formation of pulmonary hypertension.After our weeks, the pulmonary arterial pressure, cardiac output, pulmonary vascular resistance and right ventricle hypertrophy were measured.Results The mortality rate in high dose group was higher than that in low - dose group and control group.Mean pulmonary arterial pressure and right ventricular wall thickness in high doses group was higher than that in moderate dose group, low dose group, and control group( P 〈0.001).Cardiac output in high doses group was lower than that in moderate dose group, low dose group, and control group( P 〈0.001).Conclusion The moderate dose monocrotaline induced pulmonary hypertension is closer to the ideal value model.
