摘要
目的 :观察芩莲正积方对Lewis肺癌小鼠转移的抑制作用,并探讨其机制。方法 :建立Lewis肺癌小鼠模型,芩莲正积方干预,并设对照组,观察各组抑瘤率、肺转移抑制率,采用SABC免疫组化法检测各组CD44含量、实时荧光定量PCR检测各组血管内皮生长因子(vascular endothelial growth factor,VEGF)m RNA表达情况。结果:芩莲正积方组、顺铂组及联合组的抑瘤率分别为18.16%、27.12%、36.37%,芩莲正积方可抑制肺癌转移,降低CD44、VEGF表达,与顺铂联用作用增强。结论:芩莲正积方可抑制Lewis肺癌小鼠转移,其机制可能与降低CD44、VEGF表达相关。
Objective:To observe the effects of Qinlian Zhengji formula on inhibitiing metastasis of Lewis lung cancer mouse,and to discuss its related mechanisms. Methods:Models of lewis lung cancer mouse were established for different groups of intervention. To observe groups of tumor inhibition rate and lung metasta- sis inhibition rate,test groups of CD44 content by immunohistochemical,use real-time fluorescent quantitative PCR technique to detect the expression of VEGF gene. Results:The tumor inhibition rates of Qinlian Zhengji formula group,cisplatin group and combined group were 18.16%,27.12%,36.37%,Qinlian Zhengji formula can inhibit lung cancer metastases,and reduce the expression of CD44 and vascular endothelial growth factor (VEGF) gene expression. It combined cisplatin has synergistic effect. Conclusion:Qinlian Zhengji formula can inhibit metastasis of Lewislung cancer mouse,its mechanisms may be related to its decreasing the expression of CD44 and VEGF.
