摘要
本实验采用单因素及正交试验对微波法辅提灵芝多糖提取工艺条件进行优化,以抗坏血酸为阳性对照对灵芝粗多糖体外抗氧化活性进行评价.实验结果显示灵芝经蒸馏水预浸泡1h后在温度60℃、料水比1∶40、微波功率100 W、微波时间30min的提取条件下多糖得率最高.灵芝粗多糖体外抗氧化活性试验证明灵芝粗多糖有一定的体外抗氧化活性且与浓度呈正相关,但总体不及抗坏血酸.当灵芝多糖浓度为2 mg/mL时,对·OH和O2-·的清除能力最大分别为62.07%和55.39%,说明其对活性氧自由基的清除具有选择性,可能与其粗多糖中的杂质、多糖结构和清除机理有关.
In this paper ,single factor and orthogonal experiments were designed to optimize the microwave extraction conditions of ganoderma lucidum polysaccharides from the aspects of temperature ,material‐water ratio ,microwave power ,microwave extraction time in this pa‐per .Results showed that after pre‐soaking in distilled water for an hour ,when the tempera‐ture ,material‐water ratio ,microwave power and microwave extraction time was respectively 60 ℃ ,1∶40 ,100 W ,30 min ,the obtaining rate of polysaccharides was the highest .It proved that ganoderma crude polysaccharides had some reducing ability and scavenging capacity of different radicals ,w hich meant it had a certain selectivity of scavenging free radical .It might be related to impurities in the crude polysaccharides ,polysaccharide structure and scavenging mechanisms .
