辽宁汉族2型糖尿病患者脂联素基因T45G多态性与吡格列酮疗效的相关性 被引量:2

摘要 目的探讨辽宁汉族2型糖尿病(T2DM)患者脂联素(ADPN)基因T45G多态性与吡格列酮疗效的相关性。方法对97例新诊断的T2DM患者进行ADPN基因T45G多态性检测后,予吡格列酮治疗12周,治疗前后分别检测血清ADPN及糖、脂代谢、胰岛素抵抗指数(HOMA-IR)等相关指标。结果突变(TG+GG)组基线体重指数(BMI)、腰臀比(WHR)、HOMA-IR高于TT组(P<0.05),ADPN低于TT组但差异无统计学意义(P>0.05)。治疗12周后,突变组ADPN水平和HOMA-IR较基线增高(P<0.01)。突变组治疗有效率(55.51%)显着高于TT组(32.69%)(P<0.05)。Logistic回归分析显示,ADPN基因SNP45T/G是影响吡格列酮治疗有效性的独立相关因素。结论 ADPN基因SNP45T/G多态性与肥胖、胰岛素(INS)敏感性、血清ap M1水平相关。ADPN基因T45G多态性影响吡格列酮疗效。 【Objective】 To explore the correlation between adiponectin(ADPN) gene T45 G polymorphism and Pioglitazone curative effect in type 2 diabetes of the Han nationality in Liaoning Province. 【Methods】Totally 97 patients newly diagnosed as type 2 diabetes were detected for T45 G polymorphism of ADPN gene.Then the patients received Pioglitazone drug treatment for 12 weeks. Serum ADPN, lipid and carbohydrate metabolism related indexes were assessed at the beginning of the treatment and after treatment. 【Results】 The baseline body mass index(BMI), waist-to-hip ratio(WHR) and insulin resistance index(HOMA-IR) in the mutation(TG+GG) group were significantly higher than those in the TT group(P 〈0.05), while ADPN was insignificantly lower than that of the TT group(P 〉0.05). After 12 weeks of treatment, the ADPN and HOMAIR levels in the mutation group increased compared to the baseline levels(P 〈0.01). The treatment efficiency in the mutation(TG+GG) group was higher than that in the TT group(55.51% vs 32.69%, P 〈0.05). Logistic regression analysis showed that ADPN gene SNP45T/G was an independent associated factor for the effectiveness of Pioglitazone treatment. 【Conclusions】 ADPN gene SNP45T/G is associated with obesity, insulin sensitivity and ap M1 level. ADPN gene T45 G polymorphism influences the curative effect of Pioglitazone.
