摘要
目的研究哺乳期大鼠牙周组织中血管内皮生长因子(VEGF)在哺乳不同阶段的表达,探讨哺乳对VEGF表达的影响。方法选择50只成年雌性Wistar大鼠,随机抽取5只作为正常对照组,剩下45只建立哺乳期大鼠正畸牙移动动物模型,随机分成哺乳加力组、正常加力组、哺乳不加力组,每组15只。每组按哺乳或加力7、14、21 d时间段又均分为3组,每小组各5只。于各时间段的最后一天将各组大鼠处死,取静脉血采用电化学免疫分析法检测血清雌二醇水平;取大鼠上颌第一磨牙及附近牙槽骨组织块,进行HE染色和免疫组化染色,应用半定量分析牙周组织中VEGF的表达。结果哺乳能引起大鼠血清雌激素水平下降,加力能引起哺乳期及正常大鼠雌激素水平升高。哺乳加力组大鼠在不同哺乳时间VEGF表达水平均高于哺乳不加力组和正常对照组,差异有统计学意义(P均<0.05)。哺乳加力组大鼠不同加力时间VEGF表达水平均高于正常加力组,差异有统计学意义(P均<0.05)。结论哺乳期大鼠正畸牙移动过程中牙周组织中VEGF的表达增高,提示哺乳期牙周组织改建活跃,临床治疗宜采用轻力。
Objective To investigate the expression of vascular endothelial growth factor (VEGF) in periodontal tissues during orthodontic tooth movement in different stages of rat lactation and explore the impact of lactation on the expression of VEGF. Methods Fifty female rats were selected in this study. Five rats were served as normal controls ( control group) ; lactation rat orthodontic tooth movement model was established in the remaining 45 rats, who were randomly divided into lactation and orthodontic force group, normal orthodontic force group and lactation without orthodontic force group ( n = 15 each) and were respectively re-divided into 7 d, 14 d and 21 d subgroups ( n = 5 each) according to lactation time or forcing time, and the rats were respectively killed after 7 days, 14 days and 21 days. The serum estradiol (E2) level was detected by electrochemiluminescence immunoassay. The tissue blocks of the first molar of maxillary and nearby alveolar bone were taken for performing HE staining and immunohistochemical staining and semi-quantitative analysis of VEGF expression in periodontal tissues. Results Lactation can lead to the reduction of serum estrogen level of rats, and orthodontie foree can lead to the inerease of serum estrogen level in lactation rats and normal rats. VEGF expression levels of different lactation times in lactation and orthodontic force group were all significantly higher than those in lactation without orthodontic force group and control group ( all P 〈 0.05 ). VEGF expression levels of different forcing times in lactation and orthodontic force group were all significantly higher than those in normal orthodontic force ( all P 〈 0.05 ). Conclusions The high-expres- sion of VEGF in periodontal tissues in the process of orthodontic tooth movement of lactation rats indicates that the perio- dontal tissue remodeling in lactation rats is active, and so light orthodontic force should be adopted in clinic therapy.
