杜仲对链脲佐菌素致糖尿病小鼠降血糖作用 被引量:7

The hypoglycemic effects and mechanism of eucommia extracts on streptozotocin induced diabetic mice

摘要 目的:研究杜仲对链脲佐菌素(STZ)致糖尿病小鼠血糖的影响及其作用机制。方法:50只小鼠尾静脉注射STZ 120mg/kg,尾静脉注射前禁食12 h。尾静脉注射72 h后检测小鼠的空腹血糖(FBG),FBG≥11.1 mol/L则说明糖尿病小鼠模型造模成功。将成模的小鼠随机分为:模型组、罗格列酮组(4 mg/kg)、杜仲水提物10 g/kg、5 g/kg、2.5 g/kg组,另设10只昆明小鼠为正常组。连续灌胃给药14天,每天1次,正常组和模型组每日灌胃给与等体积的生理盐水。末次给药前禁食12小时,给药1小时后拔眼球取血,离心取血清待测。给药后14天检测小鼠空腹血糖(FBG);对小鼠胰腺组织进行病理学观察;放射免疫测定法测定小鼠血清中空腹胰岛素(FNS)含量,计算胰岛素敏感指数(ISI),试剂盒测定血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPx)的活性和丙二醛(MDA)的含量;蛋白免疫印迹法测定胰腺组织中含半胱氨酸的天冬氨酸蛋白水解酶-3(caspase-3)、含半胱氨酸的天冬氨酸蛋白水解酶-7(caspase-7)的蛋白含量。结果:与模型组比较,杜仲水提物10 g/kg给药组的小鼠血糖(12.08±2.65)显著降低,胰岛素敏感指数(15.25±3.58)上升;血清中SOD(255.42±26.26)和GSH-Px(598.24±79.57)活性升高,MDA(9.25±2.86)含量有所下降;胰腺组织中caspase-3、caspase-7的蛋白含量(0.31±0.09,0.42±0.23)下降。结论:杜仲水提物对糖尿病小鼠具有降血糖作用,其作用机制可能与提高小鼠机体内的抗氧化能力,以及降低胰腺中caspase-3、caspase-7的蛋白含量相关。 Objective: To investigate the hypoglycemic effects and mechanism of Eucommia extracts on streptozotocin induced diabetic mice. Methods: Fifty SPF 12 hours fasted Kunming mice were injected with 120 mg / kg streptozotocin( STZ) via tail vein. Seventy-two hours after tail vein injection,the fasting blood glucose( FBG) of mice were measured,mice with FBG≥11. 1 mol / L were treated as diabetes. Diabetic mice were randomly assigned into following groups: model group,rosiglitazone group( 4 mg / kg),10 g / kg,5g / kg,2. 5g / kg dose Eucommia extracts groups; healthy mice were treated as normal group. Each group was consisted of 10 animals. Mice were intragastrically administered once a day for 14 days. Normal and model groups were given equal volume of saline. The mice were fasted overnight before the last administration,1 h after last administration mice were executed. Blood samples were collected and centrifuged to serum. The FBG were measured at14 th day; pathological changes of pancreas tissues were observed; the serum fasting insulin( FNS) was measured using radioimmunoassay and then calculated the insulin sensitivity index( ISI); superoxide dismutase( SOD),glutathione peroxidase( GSH-Px) and malonaldehyde( MDA) serum levels were detected. The caspase-3,caspase-7 protein expressions of pancreas were analyzed by western blot. Results: Compared with model group,the blood glucose of 10 g / kg dose Eucommia extracts group was significantly decreased( P〈0. 05),ISI was increased( P〈0. 05); activities of SOD and GSH-Px were risen and MDA level was decreased( P〈0. 05); the pancreas caspase-3,caspase-7 protein expressions were down regulated( P〈0. 05). Conclusion: Eucommia extracts exhibited hyperglycemic effects on diabetic mice,the involved mechanism potentially correlated with enhancing anti-oxidant capacity and lowering the protein levels of caspase-3,caspase-7 in pancreas.
