摘要
为建立黔产辣蓼的薄层鉴别与含量测定方法,采用薄层色谱法,以槲皮素为对照,以甲苯-甲酸乙酯-甲酸(10∶8∶1)为展开剂,建立了其定性鉴别方法;采用高效液相色谱法,以槲皮素为对照,Hypersil ODS2C18(5μm,200mm×4.6mm)为色谱柱,0.20%磷酸溶液-乙腈(80∶20)作为流动相,二极管矩阵检测器为检测器,建立了其含量测定方法。结果表明:建立的TLC法能很好分离各成分,其方法学考察精密度、重复性、稳定性和加样回收率的RSD分别为0.18%、0.71%、1.10%和2.44%,能够准确测定辣蓼中槲皮素的含量。
The TLC identification method of quercetin is established by using toluene-ethyl formate formic acid(10 : 8: 1)as a developing solvent and taking quercetin as CK. The content of quercetin was determined by HPLC under the conditions including chromatographic column of Hypersil ODS2 C18 (5 μm,200mm×4.6mm), moving phase of 0.2% phosphate-acetonitrile (80 : 20) and photodiode array detector to establish TLC identification and content determination method of quercetin in P. hydropiper from Guizhou. Results.The established TLC method had a good separation effect on each component. The RSD of accuracy, repeatability, stability and recovery of the established HPLC method is 0. 18%, 0. 71%, 1.10 %and 2.44 %respectively, which can determine the content of quercetin in P. hydropiper accurately.
