摘要
目的:通过观察加味麻杏石甘汤对放射后肺泡Ⅱ型上皮细胞(AECⅡ)转化生长因子-β1(TGF-β1)m RNA、Smads表达的差异,探讨该方抗放射性肺损伤的分子机制。方法:清洁级SD大鼠18只,加味麻杏石甘汤灌胃给药3d,末次给药后1-2h采血,制备含药血清。用3.64Gy/min,总剂量8Gy的X线照射小鼠AECⅡ细胞,建立放射性损伤的AECⅡ细胞模型。RT-PCR法检测AECⅡTGF-β1、Smads m RNA水平;Western blot法检测AECⅡSmads蛋白的表达水平。结果:与正常大鼠血清组比较,含药血清组在干预48h后,AECⅡ细胞TGF-β1 m RNA及Smad2 m RNA表达降低(P<0.05),Smad6、Smad7 m RNA表达增高;10%、15%含药血清组p-Smad2、p-Smad3蛋白表达下调(P<0.05),Smad6和Smad7蛋白的表达增高(P<0.05)。结论:调控TGF-β1/Smad信号通路可能是加味麻杏石甘汤抑制放射性肺损伤的机制。
Objective: To study the influence of modified Maxing Shigan Decoction(MXSGD) on expressions of TGF-β1 mRNA and Smads mRNA in alveolar epithelial cell Ⅱ(AEC Ⅱ) of model with acute radiation lung injury, and to explore its mechanism. Methods: Eighteen SD rats were given MXSGD by gavage twice a day for three days. The medicated serum was extracted after one or two hours of the last medicine administration. The AEC I injury model was established by X-ray irradiation, 3.64Gy/min and with 8Gy doses of X-ray. The expressions of TGF-β1 mRNA and Smads mRNA in AEC I were detected by RT-PCR. The expression of Smads protein in AEC II was detected by Western blot. Results: Compared with normal serum group, the expressions of TGF-β1 mRNA and Smad2 mRNA in medicated serum group were decreased after intervention for 48 hours(P〈0.05), however, the expression of Smad6 and Smad7 mRNA in medicated serum group increased, the expression of p-Smad2 and p-Smad3 in medicated serum medium and high dose groups were all significantly decreased(P〈0.05), while the expression of Smad6 and Smad7 mRNA in medicated serum medium and high dose groups all increased(P〈0.05). Conclusion: Modified Maxing Shigan Decoction can inhibit the acute radiation lung injury, which may be related to regulate the TGF-β1/Smad signal pathway.
