摘要
目的研究多糖对抽薹防风(出现地上茎时采集的防风)药效及其药动学的影响。方法以解热、镇痛和抗炎3项指标考察抽薹防风和抽薹防风+多糖两组对雄性SD大鼠药效强度差异,并探讨其机理。结果 "抽薹防风+多糖"各剂量组均能够使2,4-二硝基苯酚致热的大鼠体温在给药2 h后明显降低(P<0.05),并持续到给药后4 h(P<0.05),而抽薹防风各剂量组无明显的降温效果(P>0.05);与空白对照组比较,抽薹防风起效快但维持时间短,抽薹防风+多糖组持续时间长镇痛作用更为明显;与模型对照组相比,抽薹防风+多糖组对角叉菜诱导的大鼠足肿抗炎作用效果优于抽薹防风组(P<0.05)。抽薹防风+多糖使大鼠体内升麻素含有量显著高于抽薹防风,两组比较,差异有统计学意义(P<0.01)。结论多糖在升麻素苷和5-O-甲基维斯阿米醇苷向升麻素转化过程中发挥了重要作用。
AIM To explore the influence of polysaccharides on the pharmacodynamics and pharmacokinetics of bolting Saposhnikoviae Radix( Saposhnikovia divaricata,picked in appearance of aerial stem). METHODS Three indicators( antipyretic,analgesic and anti-inflammatory action) were examined for the study on the pharmacodynamics difference of bolting Saposhnikoviae Radix and bolting Saposhnikoviae Radix + pdysaccharides for male SD rats and its possible mechanism of action. RESULTS The fever induced by 2,4-dinitrophenol was reduced by bolting Saposhnikoviae Radix + polysaccharides at 2 h after the administration( P〈0. 05),and the antipyretic effect lasted for 4 h after administration( P〉0. 05),while bolting Saposhnikoviae Radix( without polysaccharides)demonstrated no significant cooling effect( P〈0. 05). Compared with the control group,bolting Saposhnikoviae Radix displayed both a short response time and a short action duration,while bolting Saposhnikoviae Radix + polysaccharides presented a rapid and prolonged analgesic power. And bolting Saposhnikoviae Radix + polysaccharides was much stronger in relieving rats' paw edema induced by carrageenan than bolting Saposhnikoviae Radix( P〈0. 05). The former brought a higher plasma cimicifugoside concentration than the latter with a significant statistical difference( P〈0. 01). CONCLUSION Polysaccharides play an important role in converting prim-O-glucosylcimifugin and 5-O-methylvisammioside into cimicifugoside.
