摘要
目的:观察桃红四物汤对早期骨筋膜室综合征模型家兔血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、谷草转氨酶(AST)、丙二醛(MDA)浓度和超氧化物歧化酶(SOD)含量的影响,为桃红四物汤的临床应用提供实验依据。方法:本实验选用30只健康日本大耳成年白兔,随机分为3组,10只为空白对照组,余20只参照改良"Aydin F止血带法"复制兔小腿骨筋膜室综合征模型,造模成功后随机分为桃红四物汤组(10只)和模型组(10只),桃红四物汤组予桃红四物汤灌胃(2.8 g/kg),模型组予等剂量生理盐水灌胃。分别于实验前、实验后2 h、24 h、48 h、6 d、1周共6个时间点连续动态采兔耳缘静脉血,检测CK、LDH、AST、SOD和MDA活性;实验结束后立即处死所有动物,取健侧和伤侧小腿肌肉进行病理切片检查。结果:造模后与造模前比较,兔伤侧小腿肿胀程度明显,实验后2 h,桃红四物汤组和模型组血中CK、LDH、AST和MDA水平均高于空白对照组,SOD低于空白对照组,差异均有统计学意义(P<0.01);实验后24 h至1周桃红四物汤组血中CK、LDH、AST和MDA水平各时间点均低于模型组,SOD水平各时间点均高于模型组,差异均有统计学意义(P<0.01);光镜检查结果显示:桃红四物汤组组织结构改变较模型组轻。结论:桃红四物汤能上调早期骨筋膜室综合征模型家兔血清SOD含量,抑制氧自由基的过度生成,降低血中CK、LDH、AST含量,从而改善细胞损伤。
Objective: To explore the effects of Taohong Siwu Tang (桃红四物汤,THSWT) on serum CK, LDH, AST, SOD and MDA in rabbit with early osteofascial compartment syndrome, and to provide experimental basis for the clinical ap- plication. Methods: The 30 healthy Japanese big ear white rabbits were selected and randomly divided into 3 groups, 10 in blank group and 20 were created as osteofascial compartment syndrome model in lower leg according to "AydinF Tourniquet method"; the rabbits with OCS were randomly divided into THSWT group (10 rabbits) and OCS model group (10 rabbits); the rabbits were given THSWT with intragastric administration in THSWT group and equal dose of saline in OCS model group. The CK, LDH, AST, SOD and MDA in Rabbit ear vein blood were tested before the experiment, as well as 2 h, 24 h, 48 h, 3 days and 1 week latter. Results: Compared with the condition before the model creation, the hurt legs were swelling obvi- ously after the experiment; the THSWT and OCS model groups showed higher levels of CK, LDH, AST and MDA than blank group, as well as lower level of SOD than the blank group, with statistical difference (P〈0.01); the THSWT group showed low- er levels of CK, LDH, AST and MDA than OCS model group, as well as higher level of SOD than the OCS model group, with statistical difference (P〈0.01); Light microscopic examination showed that the histological changes in THSWT group was lower than the OCS model group. Conclusion: THSWT can increase the serous specimen of SOD in rabbit with early osteofas- cial compartment syndrome, inhibit excessive generation of oxygen free radicals, and reduce the serous specimen of CK, LDH and AST.
