摘要
目的观察细胞凋亡与胰岛素样生长因子-1/磷脂酰肌醇-3激酶/丝苏氨酸激酶(IGF-1/PI3K/Akt)通路在糖尿病胃轻瘫大鼠胃平滑肌细胞中的变化,探讨其在糖尿病胃轻瘫发生过程中的意义。方法制备糖尿病大鼠模型,分为正常对照组(NC)组、模型2周(DM2W)组、模型4周(DM4W)组、模型6周(DM6W)组和模型8周(DM8W)组。通过观察模型大鼠胃内色素残留率和小肠传输率确定糖尿病胃轻瘫模型建立;流式细胞术检测各模型组胃平滑肌细胞凋亡率;Western blot检测各组胃平滑肌IGF-1、PI3K、p-Akt蛋白的表达。结果细胞凋亡率在DM2W、DM4W、DM6W和DM8W组逐步增高(P<0.05或P<0.01)。IGF-1、PI3K、p-AKT蛋白表达逐步降低(P<0.05或P<0.01)。结论细胞凋亡与IGF-1/PI3K/Akt通路的动态变化在糖尿病胃轻瘫发生中可能有重要作用。
Objective To observe the changes of apoptosis and IGF-1/PI3K/Akt pathway in gastric smooth muscle in diabetic gastroparesis rat and to explore the role and significance of them in the development of diabetic gastroparesis. Methods A total of 40 diabetic SD rat models were established and divided into four groups:model for two weeks(DM2W)group(n=10),model for four weeks(DM4W)group(n=10),model for six weeks(DM6W)group(n=10)and model for eight weeks(DM8W)group(n=10),meanwhile,another 10 male adult SD rats were selected as the normal control.Available diabetic gastroparesis models were confirmed by residual rate of pigment in stomach and intestinal transit rate.Apoptosis rate of gastric smooth muscle cells in each model group was detected by flow cytometry.Protein expression levels of IGF-1,PI3 Kand p-Akt in gastric smooth muscle cells in each group were detected by Western blot. Results The apoptosis rates in model groups gradually increased and protein expression levels of IGF-1,PI3 Kand p-Akt gradually decreased with model time prolonged(P〈0.05 or P〈0.01). Conclusion The dynamic changes of apoptosis and IGF-1/PI3K/Akt pathway may play an important role in the development of diabetic gastroparesis.
