摘要
为了对进境口岸截获的俄罗斯木材中的墨天牛属(非中国种)害虫进行准确、快速鉴定,笔者利用DNA条码试剂盒检测技术,基于墨天牛线粒体COⅠ(线粒体细胞色素氧化酶亚基I)基因设计一对巢式引物,对截获墨天牛属中的8个种进行COⅠ扩增,将扩增序列上传至有害生物信息网站,依据碱基位点在不同种间的排列和组成的差异,获得不同种在相应位点上的特有碱基,即标记性碱基,以此作为区分墨天牛种的依据。通过这种墨天牛属DNA条码试剂盒检测技术,可为口岸检疫鉴定提供有效的技术手段。
In order to rapidly and accurately identify theMonochamusof wood from Russian, we employed DNA barcodedetection kit technology and designed a pair of nested primers based on mitochondrialCOⅠ gene(mitochondrial cyto-chrome oxidase subunit I). Furthermore,COⅠ gene sequence of eightMonocharnusspecies was amplificated, and up-loaded to pest information site. According to differences of arrangement and composition among these species at basesites, species unique bases was got in the corresponding sites, which is named specific site to distinguish this species. Inthis way, we developed the DNA barcoding identification kit technology toMonochamusspecies, which will provide anew way for port quarantine identification.
